2018-01-012024-05-15https://scholars.lib.ntu.edu.tw/handle/123456789/664477摘要:隨著社會經濟的發展,伴侶動物的飼養不但十分普遍,而且如同家人一般被當作家庭成員一份子的情況也越來越被常見,這種變化對於工業化社會中常見的核心家庭,特別是子女成長後的空巢期核心家庭,提供豐富與穩定的情感慰藉,有益於社會發展。然而隨著動物醫療的進步,伴侶動物與人共同享受富足生活的同時,也與畜主一同衰老,與熟齡人士一般,年長的伴侶動物對於營養保健食品的需求也在逐年上升,以因應隨著老化而增加的慢性病比例。為了協助國人對健康食品把關,衛生署近年已針對健康食品的功效認證,建立了一系列保健食品功效認證之機制以及健康食品標章之審核發放系統。然而,國內對於動物健康食品之認證審核系統仍然相當欠缺。此外,一般家畜動物之飼料添加物之篩選,在試驗時,皆使用哺乳動物,如:小鼠,作為篩選試驗模型。然而,全球動保團體對於動物試驗的看法,皆有減量、替代與精緻化之要求,也因此,利用非哺乳動物之脊椎動物替代哺乳動物模型,成為重要的發展趨勢。本計畫擬以斑馬魚為模型建立非哺乳動物之飼料添加物有效成分篩選平臺,以符合動物保護與實驗動物之全球發展趨勢,同時建立動物健康食品與飼料添加物之認證審核機制,以維護動物健康食品市場之健全發展。在本年度的計畫中,我們將以斑馬魚建立健康減重之模型,以作為功效驗證與有效成分之快速篩選平台。為達成此一目的,本年度首先擬利用斑馬魚性喜逆流之特性,設計一斑馬魚之運動裝置,其次將利用本裝置建立運動對斑馬魚體重、體長、骨質密度、體組成變化等各種生理影響之觀測模型。隨後即可利用此一運動模型作為正對照組,為篩選控制動物體重用之健康食品或飼料添加物之有效成分,建立一篩選認證平臺。本年度之具體目標為: (一)設計一項標準化之斑馬魚運動裝置; (二)建立運動對斑馬魚生理影響之觀測模型; (三)以斑馬魚模型建立體重控制相關健康食品或飼料添加物之有效成分篩選認證平臺。 達成本年度目標後,在學術成就上,將會有論文發表在國際上重要期刊至少 1 篇以及國際學術研討會至少 1 篇;在技術創新成就方面,我們將建立並提供「以斑馬魚模型進行動物健康食品與飼料添加物之篩選與驗證」之服務項目;在社會影響力方面,我們將試圖開發新的商品,並提升經濟產業服務之水準;在非研究類的成就方面,將培育 2 名碩士畢業生,以為將來學術或產業研究之人力儲備。<br> Abstract: With the social and economic growth, the companion animals are now widely accepted as intimate family members. These companion animals provide a great emotional support in the modern nuclear family, especially during the adjustment of empty nest, and hence are beneficial to the entire society. However, as the progression of veterinary medicine, the average life spans of companion animals are growing longer as their human owners, the need for the healthy diet and dietary supplement for aging is also growing. To protect the consumers, the Taiwan Food and Drug Administration (TFDA) established validation methods and a certification system for functional foods and supplements. However, this type of system remains absent for animal feed and additives. On the other hand, this type of functional validation usually use mammals as models according to the published methods by TFDA, while the a global trend to Reduce, Refine and Replace the mammalian animal models is emerging. To fit in the global trend of mammalian welfare and to begin to establish validation system for animal healthy food and additives, we aim to establish a zebrafish model for body weight control. To this end, we will design a device for zebrafish exercise (swimming), and monitor the physiological effect of exercise to adult zebrafish, so that we can use zebrafish to model healthy body weight control.
