2010-04-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/684798摘要:本計畫將利用生長試驗、體外培養與生理生化指標評估抗生素取代物對肉雞腸健康狀態之影響,同時釐清瘤胃細菌素及乳酸菌間之交互作用。對生長性能與腸道健康之關係,將評估飼料原料組成和抗生素取代物對白肉雞生長性能、免疫能力發育及營養份吸收運輸之影響。試驗選用六&#32957;黴素,配合靈芝發酵產物及開發自瘤胃細菌素為抗生素取代物,添加於飼糧中,針對白肉雞生長性能、腸道先天免疫與後天免疫反應、腸道營養份吸收與運輸及血液生化指標之表現等方面進行探討。經由腸細胞體外培養測試抗生素及其替代物對腸道營養吸收及免疫之影響,評估相關分子生物標記的可行性。透過體外發酵之檢測模式,藉由添加特定益生菌與細菌素,利用測定發酵液中菌相變化及乳酸菌種類分析,來釐清細菌素及乳酸菌間之交互作用。於離體共同培養具優勢族群的乳酸桿菌與指標性細菌(腸球菌),於添加細菌數粗萃取液後,比較兩者在生長上的相對改變,釐清細菌數可能作用的乳酸桿菌族群。上述工作將可建立抗生素及其取代物對白肉雞腸道生理指標之影響。<br> Abstract: This project attempts to elucidate the effects of bacteriocin and probiotic on the boriler’s gut health, and clarify the relations between the ruminal bacteriocin and Lactobacillus. We will evaluate the effects of various ingredient compositons and antibiotic replacement on the growth performance, nutrient absorption and transportation, and immunal development in the intestinal of the broiler. Nosiheptide as the antiobiotics positive control, bacteriocin and Ganoderma lucidum polysaccharides will be supplemented in the broiler feed, and study the response of immune response, nutrient absorption and blood biochemical assay. These parameters used above will be evaluated as the potential biomarkers for antiobiotic replacement. The in vitro fermentation technique will apply on this process to assay the influence of different level bacteriocin and specific probiotics addition on gut flora. The stimulation effect of bacteriocin on probiotic performance will be identified through 16s rDNA method and test suitable probiotic bacteria strain for bacteriocin addition. The best combination of bacteriocin and probiotic mixture will evaluate with broiler feeding experiment. The effects of bacteriocin on the growth of Lactobacillus and indicator bacteria (Enterococcus) in vitro will be studied in this study. We will screen a target Enterococcus, measure the rate of growth for Lactobacillus and Enterococcus with/without bacteriocin in vitro.
