2014-08-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/691642摘要:根據2008年的國民營養調查顯示,台灣地區總體肥胖盛行率高達17%。此外,肥胖常伴隨代謝症候群的發生,而代謝謝症候群是心血管疾病主要的危險因子。台灣因為治療心血管相關疾病,更造成每年高達兩百一十六億台幣的醫療耗損。因此若能了解代謝症候群引起心血管疾病的致病機轉,不僅能維護國人健康,更有助於減少國家的醫療支出。 先前研究發現透過活化自我吞噬機制(autophagy),可減少細胞凋亡,增進衰竭心臟的能量供應,並改善心臟收縮功能。提升脂締素及其受體的表現則可減少因肥胖引起的心肌肥大。據此,我們合理推論:代謝症候群引發的心血管損傷可藉由啟動autophagy與脂締素而改善。 本計劃預計達到的目標 1 利用飲食引起肥胖的李宋豬模式,探討代謝症候群對心血管疾病預後的影響 2 釐清代謝症候群增加心血管疾病的可能機制,並進一步研究autophagy與脂締素在其中所扮演的角色 本研究的結果將可釐清代謝症候群增加心血管疾病的機制,並說明autophagy,脂締素與心血管疾病的關連,進而調控心血管疾病的預後。 <br> Abstract: According to National Nutrition Survey 2008, the prevalence of obesity in Taiwan is 17%. Metabolic syndrome, highly associated with obesity, is one of the major risk factors of cardiovascular diseases. In addition, the annual medical cost of the cardiovascular diseases in Taiwan is 216 billions NT dollars. Therefore, to elucidate the mechanisms of cardiovascular diseases induced by metabolic syndrome not only improve the public health but also decrease the medical cost. Previous study demonstrated that activating autophagy decreased the cardiomyocyte apoptosis, enhanced the energy supply of failing heart, and therefore improved the contractile functions. Increasing the expression of adiponectin and its receptor restored the myocardio-hypertrophy induced by obesity. Accordingly, we presume that activating autophagy and adiponectin improves the cardiovascular damage induced by metabolic syndrome. The major goals for this project are: 1. To clarify the prognostic significance of the metabolic syndrome for cardiovascular outcomes by using dietary-induced obese Lee-Sung miniature pig model 2. To elucidate the mechanisms by which the metabolic syndrome increases cardiovascular risk, and the role of autophagy and adiponectin on the mechanism of MetS will be further investigated The results of this study will elucidate the mechanism of metabolic syndrome-induced cardiovascular diseases, clarify the relationship among autophagy, Adiponectin and cardiovascular diseases, thereafter might regulate the progression of cardiovascular diseases.代謝症候群心血管疾病李宋豬metabolic syndromecardiovascular diseaseLee-Sung- miniature pigs利用李宋豬探討代謝症候群與心血管疾病的關聯與機制
