2012-08-012024-05-14https://scholars.lib.ntu.edu.tw/handle/123456789/658321摘要:非酒精性脂肪肝可以是單純的肝臟脂肪浸潤,但在長時間或大量的脂肪堆積會導致肝細胞發炎反應,可能進一步造成肝細胞壞死、纖維化、肝硬化等一連串的肝病變。由於患有非酒精性脂肪肝的人相當多,因此草藥或保健食品在預防非酒精性脂肪肝及避免發展成脂肪肝炎及肝纖維化有相當的發展空間。這個計劃我們擬從先前研發的一個可有效預防非酒精性脂肪肝、脂肪肝炎及肝纖維化的草藥配方中尋找出有效成分,並探討是否有治療脂肪肝炎及肝纖維化的可能。這個配方是由3 個草藥組合而成,我們擬以單一藥材或是兩兩組和進行動物實驗的方式找出較具有效果的藥材,再進行有效成分的分離及鑑定。有效成分將以能夠抑制肝細胞三酸甘油脂含量與肝臟星狀細胞活化的為目標,之後再以動物實驗驗證其預防及治療效果。這個計劃採用的動物模式是我們最近發展出來,單純以高脂高膽固醇飼料引起非酒精性脂肪肝炎及肝纖維化的動物,不同於以往的模式都是以化學藥劑引起肝炎,初步實驗也顯示我們的草藥配方有顯著預防非酒精性脂肪肝炎及肝纖維化的效果,因此只要能找到有效成分將有利於專利申請與產學合作之進行。<br> Abstract: Non-alcoholic fatty liver disease (NAFLD) can be simple liver steatosis;however, approximately 30% NAFLD were found to have an inflammatoryphenotype referred to as non-alcoholic steatohepatitis (NASH) which is believedas a major cause of fibrosis and cirrhosis of the liver of unknown cause. BecauseNAFLD is increasing in prevalence worldwide; therefore, market for preventionand treatment of NAFLD has great potential. Herb medicine has beendemonstrated to exhibit therapeutic potential to many diseases. We havedeveloped a new herbal formula that is potent in prevention of NASH andfibrosis in two diet-induced NAFLD animal models. In this grand proposal, weaim to identify and isolation of the active components from the herbal mixture.Moreover, the therapeutic potential for NASH and liver fibrosis will beevaluated. The formula is composed by 3 herbs. First, we will investigate theeffect of single herb or combination of 2 herbs on high fat/cholesteroldiet-induced NASH and liver fibrosis. Then isolate effective compound that caninhibit fat accumulation in hepatocytes and inhibit activation of hepatic stellatecell from a single effective herb; or make a better combination of the formula.Then test the in vivo effect of the product. We will use a newly establishedunique animal model that develops NASH and liver fibrosis by highfat/cholesterol diet without any chemical inducer. Our preliminary results showthat the herbal mixture is potent in prevention of NASH and fibrosis in thisanimal model.
