2020-06-012024-05-18https://scholars.lib.ntu.edu.tw/handle/123456789/700093摘要:隨著全球人口老化問題,人類的平均壽命增加,全球醫療機構統計之器官捐助的需 求也逐漸上。3D生物列印,是一種將生物相容性材料與細胞混和後共同列印的精 密技術,可客製化製備成與天然組織和器官相似的人造替代物,此人造替代物可替 換至受傷的組織和器官進行修復、重建或作為藥物篩選的平台。目前市面上雖已有 多種3D列印生物墨水,但材料的使用上仍有許多限制存在,無法利用一種簡單的 生物墨水列印不同的細胞。本二年期產學計畫主要為設計與研發生物可降解聚胺酯 作為3D列印生物墨水材料之系列產品。生物可降解的聚合物生物墨水需具備生物 相容性及生物可降解性外,亦需良好的機械性質以確保混合細胞列印時,能維持完 整支撐強度。生物可降解聚胺酯生物墨水材料,可經過嚴謹標準合成流程及製程設 計來建立符合需求及保存的環境。本計畫第一年擬發展模組化的製程,系統化的建 立應用於不同組織和器官之墨水配方,並分析其降解速率及機械強度,建立對於各 組織和器官最合適之生物可降解聚胺酯生物墨水配方;第二年擬優化生物可降解聚 胺酯生物列印墨水製程,並加入不同來源的細胞進行列印,評估不同生物墨水配方 之細胞與生物相容性。透過此二年期產學合作計畫,期望能完成生物列印墨水產品 化之開發與評估,以強化合作雙方未來共同開拓生物可降解聚胺酯生物列印墨水商 品化之目標與方向,扶植本土優良傳統產業進行高附加價值醫療器材原料與產品之 開發與製造。<br> Abstract: The world`s population is aging, the average human life expectancy has increased, and the demand for organ donation estimated by global medical institutions has gradually increased. The 3D bioprinting is an emerging technique to create tissue-like structures for tissue or organ repair, reconstruction, and as a platform for drug screening. Recently there are some formulae of 3D printing bioink designed for various bioprinting platforms on the market. However, the existing formulae of bioink still have many restrictions to be solved (i.e. completely different ink for different cells). The focus of the project is to design and develop a series of 3D printing bioink derived from water-based biodegradable polyurethane. The series of 3D printing bioink have high biocompatibility, printability, biodegradability, and mechanical properties. In the first year of the project, we will mainly focus on systematically establish bioink formulae applied to various cell types, particularly by using a “modular” approach. We will confirm the stability, physical, chemical properties, degradation rate, and mechanical properties of bioink. In the second year, we will evaluate the biocompatibility of bioink using several types of cell lines. Furthermore, we will develop storage conditions and the shelf life of bioink.
