2008-06-012024-05-18https://scholars.lib.ntu.edu.tw/handle/123456789/706796摘要:本計劃為延續目前研究群對台灣金線連益菌生、免疫調節、降血糖與延緩破骨細胞活性研究工作,過去研發利用活性導向的策略區分金線連具有益菌生、免疫調節、降血糖與延緩破骨細胞活性區分,並對各區分進行主要成分分析。具有益菌生活性之區分含有之主要成分為-D-葡萄聚醣與-D-半乳聚醣;具有細胞免疫調節活性之區分含有之主要成分為-D-半乳聚醣、果膠多醣與甘露聚醣;具有降血糖活性之區分含有之主要成分蘋果酸與一新穎有機酸(2,5-dioxo-hexahydrofuro [3,2-b]furan- 3a,6a-dicarboxylic acid);具有延緩破骨細胞活性之區分含有之主要成分為多酚酸醣&#33527;化合物melilotoside and its isomer (hydroxycinnamic acid derivatives)。本年度會針對有效成分發展定性與定量的分析方法,做為例行性的品保技術與改進製程的確認工具,分析方法設計的初期計畫是以-D-半乳聚醣與果膠多醣的水解酵素,將多醣轉換為單元體或雙元體,再利用高效能陰離子層析法定量分析單元體與雙元體,並使用選擇性呈色法檢測,再反推原料與產品中的半乳聚醣與果膠多醣含量,同時高效能液相層析法分析新穎有機酸與多酚酸醣&#33527;化合物,上述 -D-半乳聚醣、有機酸、多酚酸醣&#33527;化合物與知名之化合物kinsenone可做為開發金線連產品的指標性成分。<br> Abstract: This proposal is an extension work of our research on prebiotic, immune modulating, glycemia lowering and osteoclast retarding activities of Taiwnese Jewel Orchid (Anoectochilus formosanus Hayata). In previous year, bioactivity-oriented fractionation was conducted to obtain prebiotic, immune modulating, glycemia lowering and osteoclast retarding fractions from Anoectochilus formosanus Hayata. The major components found in prebiotic enriched fraction were -D-glucans and -D-galactans. -D-galactans and pectic polysaccharides along with mannans were in cyto-immune modulating fraction; malic acid and a novel organic acid (2, 5-dioxo-hexahydrofuro [3, 2-] furan- 3, 6-dicarboxylic acid) were in glycemia lowering fraction; melilotoside and its isomer (hydroxycinnamic acid derivatives) were in osteoclast retarding fraction. The works for the coming year will foucus on developing qalitative and qantative analytical methods for routine qalilty assurance and cheching technique in processing improvement. The preliminary plot for analytical methods are using -D-galactans and pectic polysaccharides related enzymes to convert polymers into their monomers or dimers and using high-performance aninon-exchange chromatoragphy to analyze the content of those monomers and dimers and to obtain the content of polymer. High-performance liquid chromatography will develop to determine content of the novel organic acid and melilotoside and its derivatives.
