2011-08-012024-05-14https://scholars.lib.ntu.edu.tw/handle/123456789/658811摘要:嘉磷塞、巴拉刈及太刈為台灣常用的除草劑。台灣除草劑中毒案例主要以自殺為主。嘉磷塞中毒時對肺臟產生傷害,巴拉刈及太刈中毒時產生急性肺部傷害/急性呼吸窘迫症及肺纖維化。傳統上對除草劑中毒時的臨床治療方式為嘔吐、活性碳治療、血液透析及藥物治療,但治療後其死亡率能偏高,因此對除草劑中毒引起的急性肺部傷害/急性呼吸窘迫症須創新療法。本實驗室已建立以除草劑巴拉刈引發的急性肺部傷害/急性呼吸窘迫症的小鼠。 此外,初步實驗數據顯示間質幹源細胞,諸如人體骨隨間質幹源細胞及人體絨毛膜間質幹源細胞可降低除草劑巴拉刈引起的死亡率、回復肺部功能及減緩肺部組織的傷害。本計畫三年的目的如下: (1)証實間質幹源細胞有效的治療急性肺部傷害/急性呼吸窘迫症的小鼠。(2)探索間質幹源細胞對傷害的肺部組織修補的作用機制。(3) 建立急性肺部傷害/急性呼吸窘迫症的豬。(4)再以証實間質幹源細胞有效的治療急性肺部傷害/急性呼吸窘迫症的豬及探索間質幹源細胞對傷害的豬肺部組織修補的作用機制以及應用於臨床足跡。<br> Abstract: It has been reported that glyphosate and bipyridylium quaternary ammonium herbicides (paraquat and diquat) were the main herbicide used in Taiwan. Lung injury occurs in all case of glyphosate intoxication. Paraquat and diquat induce acute lung injury (ALI)/acute respiratory distress syndrome (ARDS) and progressive fibrosis. The conventional clinical treatment involve such as emesis, charcoal haemoperfusion, haemodialysis and pharmacological therapies remain in high motarity rate. Therefore it needs some innovative therapies for treatment of ALI/ARDS induced by glyphosate, paraquat or diquat. In our laboratories, the experimental animal model (mice) of ALI/ARDS has been established by treatment of paraquat. Futhermore, our preliminary experimental data have been shown that the treatment with a variety of mesenchymal stem cells (MSCs) such as human bone marrow-derived MSCs (hBMMSCs) and human chorion-derived mesenchymal cells (CMCs) on paraquat induced ALI/ARDS mice improved their survival rate, pulmonary function and attenuate tissue damage in the lung. The objects of our present proposal for three years research project are as follows: (1) To confirm which MSCs is the most effective to treat the ALI/ARDS mice. (2) Further exploration of the mechanism of repairment of the damaged lung tissue by MSCs.
