2013-08-012024-05-14https://scholars.lib.ntu.edu.tw/handle/123456789/658344摘要:針對環境荷爾蒙健康危害之研究,一般均著重在產前暴露之關鍵期觀察,再從公共衛生角度進行環境介質及人體暴露量研究,並觀察孩童早期之健康影響,而基礎研究則多從動物實驗探討可能的致病機轉。然而考量環境荷爾蒙之潛在生殖危害,針對發展中胎兒影響之研究仍極為缺乏,因此很難確定在眾多複雜環境荷爾蒙暴露中,來自雙酚 A (BPA) 暴露對健康效應的貢獻。本整合型計畫希望能以完整的跨領域合作為基礎,除了公共衛生之外,將臨床生殖醫學、基礎分子生物學納入,以評估 BPA對人體之健康危害。整體研究以公共衛生及流行病學為主軸,再橫向整合臨床醫學及基礎分子生物學之研究觀察,從複雜的暴露型態中驗證母體 BPA 暴露對下一代之健康效應。總體目標在完整呈現國人 BPA 暴露水準、BPA 對人體之健康影響、致病機制探討及整合驗證,提供我國政府在規劃環境荷爾蒙管理及政策優先順序,以期達到國家永續發展的參考。本子計畫執行目的在透過採樣及實驗室模擬評析影響飲用水中BPA 濃度之因子,並透過與其他子計畫之協調合作,蒐集來自其他環境介質及食物攝取之BPA暴露,進行BPA之健康風險評估。考慮現有關於水體中BPA 之研究著重於河川水體中之濃度調查,本子計畫將著重於河川水體、淨水場原水及清水(含各處理流程)、配水至家戶用水中BPA 濃度變化之相關性探討,藉以評估及探討影響飲用水中BPA 濃度之因子。本子計畫分三年執行,其分年執行重心為:第一年 - 透過採樣調查河川水體、淨水場原水及清水、配水至用戶端水中BPA 濃度之相關性。於此同時並與其他環境暴露評估子計畫合作建立國人暴露於BPA 之總暴露評估方法。第二年 - 除持續進行水質調查外,蒐集/更新暴露參數以建立微量污染物之健康風險評估方法,據以提出適用於國人之 BPA 健康風險評估方法。第二年計畫之執行將與其他子計畫之成果聯結,整合BPA 之環境檢測、生物監測、暴露評估、健康效應調查等不同子計畫之成果,以提升健康風險評估結果之價值。第三年 - 依據環境暴露評估及風險評估結果,以BPA 為例提出BPA之風險管理策略。透過第二年及第三年計畫之執行,結合不同子計畫之成果,將可提升健康風險評估結果之實務性及學術價值。本子計畫之執行將可延續歷年計畫執行成果,結合環境檢測、生物監測、健康效應調查、暴露評估、風險評估、管制策略等不同子計畫之成果,提出具體BPA風險管理策略方案以供政府施政之用。<br> Abstract: Most of the studies related to the health effects of environmental hormone focus on theobservations from the prenatal exposures. Based on public health perspectives, some otherliteratures assessed the human exposures from various environmental media and to observe thehealth effects of early childhood. Fundamental researches for the possible pathogenic mechanismswere elucidated from animal studies. However, there is only little information available consideringthe potential reproductive effects of environmental hormones. It is difficult to differentiate thehealth effects of bisphenol A (BPA) exposure from the many complex environmental hormoneexposures.Based on interdisciplinary cooperation, the objective of this integrated project is to assess thepotential health effects of BPA. Together with public health methodology, this study integratesclinical reproductive medicine and basic molecular biology to assess the hazards of BPA on humanhealth. The overall research frame follows the general principles of epidemiology and cross linkedto clinical medicine and basic molecular biology to verify the effect of maternal BPA exposure tothe health of the next generation. The overall goal of this study id to provide a complete view ofhuman BPA exposures, effects of BPA exposure to human health, assessment of pathogenicmechanism and integrated verification via clinical studies. The results of this study can be used as abasis for preparation of the policy of environmental hormone management to achieve thesustainable development of the nation.This sub-project assess the factors affect the BPA concentrations in drinking water and thehuman exposures to BPA through oral ingestion of drinking water. Water samples will be collectedfrom river water, raw water, treated water from water treatment plants and household tap water.Laboratory simulation studies will be conduced to verify the potential BPA exposures throughhousehold water consumptions. In addition, cooperation with other sub-projects will be a key toprovide other exposure routes of BPA for total exposure assessment.The yearly work of this three-year project will be:Subproject executive three years, the center of gravity of the execution of its sub-year:The first year - Through the water samplings from river water, raw water, treated water andhousehold tap water, the degradation profiles of BPA in water environment andthe potential BPA exposures through consumptions of drinking water will beassessed. In the mean time and possible exposures from other environmentalmedia (such as air) will be assessed by the other sub-projects. The data collectedfrom different sub-projects will be integrated to establish the total humanexposures of BPA.The second year - In addition to the water quality surveys, this study will collect exposureparameters for health risk assessment of trace pollutants in order to establish amethodology for BPA health risk assessment suitable for Taiwan. The executionof the second year project will also cooperate with other sub-projects tointegration the results of BPA environmental detections, biological monitoring,exposure assessment and health effects. In this way the value of the BPA healthrisk assessment results can be enhanced through the execution of this integratedproject.The third year - Based on results of the exposure assessment and risk assessment, a BPA riskmanagement strategies will be proposed. Through the execution of the secondyear and third year project, combined with the results of the other sub-projects,this project will enhance the the practical and academic value of the health riskassessment and management.The results of this project will be able to extend the outcomes of the projects conducted in pastyears.
