2010-02-272024-05-15https://scholars.lib.ntu.edu.tw/handle/123456789/662426摘要:細胞質雄不稔在雜交一代種子生產的應用非常普遍。然而因為甜椒缺乏良好且穩定的恢復系統作為種子生產時花粉親及穩定不受溫度影響的不稔系,因此細胞質雄不稔在甜椒上的應用仍然受限。亞蔬已篩選出兩個穩定的雄不稔品系(CCA 7234、CCA7235 ) 和9個甜椒恢復系(C04972,C01757, 0337-7018, 0537-7044, 0537-7045,9852-190, 9852-131,9852-133,9950-5558),但缺乏鐘型大果的甜椒穩定恢復系可供一代種子生產使用。如鐘型甜椒“Jupiter”是一個美國很常用的鐘型自交品種,將來很有潛力成為很好的恢復系,很適合拿來做為鐘型甜椒恢復系的選育材料。在預備試驗中,本團隊已確立辣椒CMS及恢復基因(Rf)穩定表現的分子標誌系統可應用在甜椒上,並鑑定出0337-7018及9852-131兩個甜椒恢復系具有可穩定傳到雜交後代的恢復基因(Rf)分子標誌(CRF-S 引子產生的PCR產物);此外,辣椒的恢復基因轉入甜椒時,也可以此分子標誌檢測。有關番椒恢復系方面的研究,一般認為辣椒恢復系較為穩定,甜椒一般有恢復系的恢復花粉不穩定的問題;因此,第一年,鐘型甜椒的恢復系育種,欲選擇具有CRF-S 分子標誌之恢復基因之辣椒9955-15為花粉親並雜交鐘型甜椒;第二到第三年完整且穩定地將辣椒的恢復基因導入鐘型甜椒中,在此回交育種的過程中,可應用此分子標誌協助篩選,減少工作量並縮短育種時程。<br> Abstract: Cytoplasmic male sterility (CMS) has been increasingly used in F1 hybrid seed production of hot pepper because of reduced production costs. Use in sweet peppers is limited due to a lack of good CMS-restorer lines to serve as pollen parents, and because CMS-sterile lines showed poor stablility under low temperatures. The stable-CMS sweet pepper lines (CCA 7234, 7235), were crossed with sweet pepper lines with good horticulture characters to identify restorer lines (C04972, C01757, 0337-7018, 0537- 7044, 0537-7045, 9852-190, 9852-131, 9852-133, 9950-5558) but theserestorer were almost long bell shape. In sweet pepper hybrid, more restorer and bell shape need be identified. Variety “Jupiter” was a successful bell shape OP variety in the USA, and will be used as a recurrent parent. A Molecule-marker was developed to test CMS-system (Gulyas, 2006). Primer CRF-S can find restorer gene “Rf” in hot pepper. This study will used CRF-S primer to select with all CMS and big bell lines, and select best parent to backcross hot restorer "9955- 15" into sweet pepper back ground. Year 2, will test back cross and CMSF1 population to reconfirm the marker’s stability. In year 3, will throng continued back crossing and marker assistant seln bell restorer sweet pepper.
