2006-01-012024-05-15https://scholars.lib.ntu.edu.tw/handle/123456789/663244摘要:目前水解角蛋白的方法以酸水解為主,酸水解成本較高且有化學藥劑殘留的考量,因此利用酵素水解應是最佳的方法。然而目前針對酵素水解角蛋白的研究不多,且以雞毛為主,豬毛之研究付諸闕如,因此本研究擬利用具有角蛋白分解能力的菌株,例如Bacillus licheniformis., Streptomyces pactum., Aspergillus spp., 以豬毛為原料分析水解產物,篩選分解能力最佳之菌株,再利用生物工程的技術將此蛋白分解酶基因分離並插入表現載體,轉型入勝任細胞,使其大量表現,最後將蛋白分解酶純化分離後,利用固定化技術固定酵素,提高其分解效率,有利於工業上的利用。本研究期能解決豬毛不易分解處理的問題,將豬毛有效利用,生產高單價之胺基酸,減少廢棄物,增加收益。<br> Abstract: Large amounts of pig bristles are constantly generated in areas of intensive pork processing. Although pig bristle contains over 90% crude protein, it has a limited use for animal nutrition due to its high content of keratin that is poorly digested by proteolytic enzymes. As a result, pig bristle is often pressure-cooked or chemically-treated to improve its protein digestibility. However, these treatments require considerable energy consumption and have possible detrimental effects on the nutritional quality of the final products. Degradation of pig bristles by protease may provide a solution.
