2006-04-262024-05-15https://scholars.lib.ntu.edu.tw/handle/123456789/661914摘要:相思樹為台灣常見的本土樹種,主要分布於台灣低海拔山地與丘陵地。目前,有關相思樹的主要功用,除了可供作為傳統薪炭材之外,亦可作為部分結構用材。此外,相思樹心材、樹皮抽出物已證實具有抗氧化活性。然而,對於相思樹葉子抽出物之抗氧化活性,截至目前為止仍缺乏深入之研究。因此,本研究擬利用管柱層析、薄層層析、高效能液相層析等技術,分離及純化相思樹葉子抽出成分,並進行抗氧化活性評估。抗氧化試驗,擬利用DPPH自由基捕捉、超氧自由基捕捉、脂質過氧化、還原力、金屬螯合能力等體外試驗(In vitro)。經活性評估所得之抗氧化活性成分,則運用質譜分析、紅外光光譜分析以及核磁共振光譜分析等光譜分析,鑑定其確切結構。本計畫如能順利完成,則可確定具抗氧化成分,甚或了解其作用機制,如此將有助於開發本土樹種加工利用之潛能,並擴展其應用領域。<br> Abstract: Acacia confusa Merr. (Leguminosae), a species indigenous to Taiwan, is widely distributed on the hills and lowlands of Taiwan. It is a traditionally important commodity used for charcoal-making and construction materials. Previous results demonstrated that the heartwood and bark extracts from A. confusa had excellent antioxidant activities. However, the potential antioxidant activities of A. confusa leaf extracts have not been studied yet to date. In this study, column chromatography (CC), thin layer chromatography (TLC) and high performance liquid chromatography (HPLC) are employed to separate and purify the A. confusa leaf extracts, then their antioxidant activities are further evaluated by various assays using a variety of in vitro model systems, including DPPH free radical and superoxide radical scavenging assays, lipid peroxidation assay, reducing power assay, metal chelating ability assay. In addition, the chemical structure of antioxidant compounds from A. confusa leaf will be identified by the analyses of mass spectroscopy, Fourier transform infrared spectroscopy (FTIR), and nuclear magnetic resonance spectroscopy (NMR), etc. The accomplishment of this project would help determine the antioxidant components in the leaf extracts of A. confusa and also elucidate their mechanisms of the inhibitory activity.
