2011-05-012024-05-18https://scholars.lib.ntu.edu.tw/handle/123456789/706865摘要:本設施計畫的主要目標為「創建一個世界級的功能性基因體研究核心,提供使用者所需的功能基因體服務,藉以厚植台灣生醫實力,提升台灣生技產業競爭力」。在這個計畫中,我們將提供最先進的功能性基因體研究平台、一站式單一窗口服務、客製化服務和整合性生物資訊與生物統計服務。藉由此”整合性功能基因體學核心實驗室”及其提供的客製化服務,研究同仁將可以完成最尖端的基因體研究並達成追求研究卓越的目的。除了先前NRPGM C5 核心實驗室已經提供的服務外,我們將提供包括下述的新服務項目,第一、基因功能喪失shRNA 基因庫分析晶片。第二、DNA 甲基化質譜儀分析。第三、高內涵細胞影像分析系統服務。第四、人類全基因甲基化晶片服務。第五、生物資訊與生物統計分析。第六、代謝體分析服務。為了增進服務的能力與核心的技術,我們在今年內亦將致力於發展下述的研究平台。第一、單(少數)細胞基因表現與基因變異分析服務。第二、高通量微陣列樣本檢測系統。第三、新世代定序儀分析系統。第四、開發特殊樣品代謝體分析條件。代謝體學被認為是下一個十年中最重要的生物科學技術。就我們所知,本核心是台灣唯一一個提供代謝體服務的國家級核心實驗室。於過去的兩年間,在台灣大學與中研院的資助下,我們已經針對各式的新一代定序系統應用建立了分析平台,包括,全基因組定序、全轉錄組定序、微小RNA定序、特定區域之定序以及新一代定序資料的分析方式。為了整合並促進維運計畫生物資訊核心實驗室間的合作,本核心與E2 和E3 生物資訊核心達成協調與分配核心使用者服務需求的共識。同時,我們與E3 生物資訊核心亦針對雲端運算與共享NGS 新知,簽署了合作意願書。在過去的九年間,本核心的服務收入有5,500 萬,發表了、100篇以上的論文,其中一半的論文發表在高影響指數的期刊,也獲得22 個專利。我們相信本核心將可以滿足國科會研究人員的迫切服務需求,協助這些研究同仁達成他們最好的表現。<br> Abstract: The main purpose of this project is to establish a world-class functional genomicresearch center which can support the state-of the-art services for researchers tostrengthen the basis of medical sciences and the competitive ability of Taiwan biotechindustry. In this proposal, we provide advanced functional genomic tools, one-stop service,customized service and integrated bioinformatics and biostatistics analysis. By accessingthis “Integrated Core Facility for Functional Genomics” and customized services,researchers are able to perform cutting edge genomic research and achieve researchexcellence. In addition to previous services in NRPGM C5 core facility, we will open newservices including 1. Array for loss of function shRNA library screening, 2. MassARRAYfor quantitative methylation analysis using the EpiTYPERTM assay, 3. High Content CellImaging and Analysis System, 4. Infinium Human Metyhlation450 Beadchip, 5.Bioinformatics and biostatistics analysis, and 6. Metabolomics analysis. To improve theservice capability and core technology, we will develop 1. Gene expression and geneticalteration in a single (a few) cell, 2. High throughput sample ArrayPlate screening system, 3.New Generation Sequencing (NGS) data analysis besides, and 4. Analytical methods formetabolome sciences. Metabolomics is considered one of the technologies that will blossomto benefit science in Nature's 2020 vision published in the beginning of the year 2010. Thisintegrated center is the first national core in metabolomics. In the past two years under NTUand Academic Sinica support, we have established the new assay platforms for wholegenomic sequencing, whole transcriptomics sequencing, small RNA sequencing, andregional sequencing as well as NGS data analysis, for the most powerful genomic tool, NGS(new generation sequencing). To integrate and collaborate the NGS bioinformatics corefacilities, we have made an agreement with E2 and E3 for allotment of NGS analysis servicerequest and made a collaboration agreement with E3 for development of cloud computingand share update NGS knowledge. In the past nine years, the service income of this core ismore than NTD 55,000,000. Core members and users of this core have published about 100articles and half of them are published in high impact factor journals. Moreover, coremembers and users of this core have obtained 22 international patents.
