2020-08-012024-05-13https://scholars.lib.ntu.edu.tw/handle/123456789/653891摘要:在常見和罕見疾病的研究中,對於端粒和端粒酶在人類疾病中的瞭解已取得了顯著進展。端粒酶在癌症生物學、衰老、以及發育生物學中扮演著十分重要的角色。然而,對端粒酶組裝、活性調節、對端粒延伸的作用以及其功能在病理條件下所受到影響依然缺乏透撤的了解。此項研究需要整合臨床醫生、基礎科學家和流行病學之間的多學科合作。在本提案中,我將研究生理和病理條件下(特別是神經退化性疾病與白血病)的端粒和端粒酶動力學。透過國際組織合作,首先,我將找出並詳細闡明新分子在端粒酶生合成和調控過程中的角色。其次,將檢查上述疾病對端粒與端粒酶功能的影響。此項提案將整合我的專業知識,我的最終目標是獲得並提供對人類端粒的功能更好理解。<br> Abstract: Studies of common and rare illnesses have already led to remarkable progress in the understanding of the function of telomere and telomerase in human disease. Telomerase plays a vital role in cancer biology, aging, as well as development biology. However, there is a profound lack of understanding of telomerase assembly, activity regulation and its action on telomere elongation. Moreover, it is still unclear how telomerase and telomere functions are affected under the pathological condition. Therefore, Multi-disciplinary collaborations between clinicians, basic scientists, and epidemiologist are essential to incorporate new telomere biology discoveries to improve health outcomes. In this proposal, I will focus on investigating telomere and telomerase dynamics under physiological and pathological conditions, especially leukemia and neurodegenerative diseases. By cooperating with several outstanding international groups, first, I will identify new players and elucidate biogenesis and regulation of telomerase in molecular detail. Second, the effect of diseases as described above on telomere/telomerase functions will be examined. This research proposal will be benefit from my expertise in telomere biology and RNA biology and provides me an opportunity to explore new fields in cancer biology and reproduction biology, while establishing various collaborations both locally and with international groups.
