國立臺灣大學農藝學系暨研究所謝兆樞2006-07-262018-07-112006-07-262018-07-112005-07-31http://ntur.lib.ntu.edu.tw//handle/246246/19850在後基因體時代,研究突變體 在了解各基因的功能上扮演著及 其重要的角色。此其中,包括了誘 導突變、研究突變性狀、釣取目的 基因以及分析特定的基因等。水稻 定序草圖已初步完成,它帶動了活 絡的功能性研究,在台灣就已經有 一些研究單位利用T-DNA tagging、Ac/Ds tagging 及sodium azide 等技術發展出三個水稻的突 變體族群。從這些族群我們能夠篩 選許多跟種實性狀相關的突變 體,在本三年的計劃中,我們將對 這些材料進行分析與利用。 在未來的三年內、我們擬著重 在下列三種跟胚乳相關的性狀:糯 性及半糯性、有色胚乳及香氣。由 於這些突變性狀都在日本型品種 所誘導,我們也將進一步將這些突 變體和印度型材料進行雜交,產生 F2 分離族群用於遺傳研究及定點 選殖的工作。 第一年我們已從T-DNA tagging sodium 及azide-indued 族 群篩選黃色胚乳得突變體;同時也 獲得碳化古米樣品可一起進行相 關的比對研究。At the post-genomics era, studies on the mutants play a central role to understand the function of each genes. This includes inducing mutants, study on mutated traits, pull out target gene through forward or reverse genetics, and perform the analysis for that specific gene. The rice genome sequencing draft was released recently by the international consortium and two private sectors. Here in Taiwan, several groups have been generating rice mutants through T-DNA tagging, Ac/Ds tagging and sodium azide treatment recently. From those populations, we are able to screen several mutants on grain composition. They would be used as the material for the present project. In the next three years, we will emphasize on the following three mutant characteristics: waxy (and so-called semi-waxy) rice, colored rice, and fragrant rice. We will also cross these mutants with indica rice for the generation of F2 segregating population. They will then be used for the material of genetic analysis and positional cloning. In the first year, we have obtained several color endosperm mutants from T-DNA tagging sodium azide-induced mutated populations.
