2008-03-282024-05-14https://scholars.lib.ntu.edu.tw/handle/123456789/657072摘要:本研究透過與包括屏東科技大學黃美秀博士在內之IUCN熊類專家群 (Bear Specialist Group)成員、東南亞動物園協會(SEAZA)、圈飼熊類的飼主等之合作,分年收集圈養與野外取得之亞洲黑熊與臺灣黑熊DNA樣本,挑選適當的分子標記,建立以分子生物技術檢測黑熊遺傳多樣性之流程,以進行黑熊三個層面的保育遺傳研究:(1)亞洲不同地理區、不同亞種亞洲黑熊族群的遺傳變異,(2)分析臺灣島內臺灣黑熊之族群遺傳結構,(3)以微衛星體基因座個體標定與Amelogenin基因座性別標定的方法研究臺灣黑熊的活動範圍、族群大小與性比,以掌握黑熊的族群結構及空間分布等資訊。 本年度工作包括1.持續蒐集國內外亞洲黑熊與臺灣野外及圈養黑熊個體之血液、毛髮或糞便樣本。 2.利用不同分子標誌,包括本研究團隊所研發的微衛星體基因座與Amelogenin基因座分析亞洲黑熊亞種間、地區間,台灣黑熊在島內不同地點的遺傳變異,並了解台灣黑熊之族群結構及空間分布狀況。 3.利用建立之亞洲黑熊亞種分子鑑定技術,判定目前眾多未知來源的圈養黑熊亞種別,以利後續的域內或域外保育。 4.繼續聯繫各地專家,維繫亞洲不同地理區、不同亞種亞洲黑熊族群遺傳變異研究的國際合作管道。 <br> Abstract: By working with members of the IUCN bear specialist group, SEAZA and bear keepers, this project intends to collect DNA samples of captive or wild Asiatic black bears, including the Formosan black bears, select appropriate genetic markers, and set up standard operation procedures of collecting, storing and analyzing bear DNA samples, so that we can: 1. examine genetic variation of Asiatic black bears from different parts of Asia; 2. analyze the genetic structure of the Formosan black bears from different parts of Taiwan; and 3. develop appropriate methodology in examining various aspects of population ecology of the Formosan black bears.
