2011-08-012024-05-13https://scholars.lib.ntu.edu.tw/handle/123456789/645109摘要:雌激素為一種與生殖內分泌有關的固醇類賀爾蒙,被認為具有保護與滋 養神經的效果。然而,使用雌激素作為停經後婦女賀爾蒙補充治療的經驗, 卻顯示雌激素可能有增加乳癌與心血管疾病的風險。包括大豆甙元等物質 的植物雌激素,廣泛存在於大豆等日常食用植物,在結構與生物活性上類似 雌激素,但較無增加疾病的風險。因此,大豆甙元倘若也有保護與滋養神 經的作用,則將有很高的臨床應用價值。背根神經節的神經元細胞在神經系統中,主司將感覺訊息自週邊組織傳 遞到中樞神經系統的脊髓,若因神經疾患或損傷而造成其功能的喪失,將導 致病患生活上的不便與後遺症。另外,在腦部與脊髓組織的星狀細胞,扮演 支持與營養神經細胞的角色,也極易在許多神經系統疾患中受傷而無法正常 作用,更增加神經系統的障礙。前述曾提到,雌激素具有保護與滋養神經的 效果。因此,大豆甙元是否有滋養與保護背根神經元與星狀細胞的作用, 係一個十分重要且有趣的課題。因此,本計畫將針對大豆甙元的神經滋養 與保護作用,與其可能的機轉,以細胞培養與脊髓損傷動物實驗來進行深入 的研究與探討。<br> Abstract: Estrogen, a steroid hormone with important endocrine function in sexual differentiation and reproduction, has been implicated to have neuroprotective and neurotrophic effects. However, the use of estrogen for hormone replacement in postmenopausal woman has raised concern about its potential risk of increasing incidence of breast cancer and cardiovascular event. Daidzein, a molecule belonging to the family of phytoestrogen which is structurally and functionally related to estrogen, may have smiliar property of neuroprotection and neurotrophism, but not estrogen’s side effects. A naturally ingredient in our daily food including soy, daidzein may be a promising agent ready for clinical use if it has beneficial effect on nervous tissue after neurological trauma or diseases.Dorsal root ganglion (DRG) neurons function to receive sensory inputs from peripheral tissue and relay the signal to the central nervous system. Dysfunction of DRG neurons due to neurological insult results in significant morbidity. Astrocytes, the supporting cells in central nervous system, could also be damaged in various insults to the brain and spinal cord. Since estrogen has both neurotrophic and neuroprotective properties, it is interesting to know whether daidzein has such effects for the DRG neurons and astrocytes. In the current research project, we aim to investigate these issues with both in vitro and in vivo approaches.
