林大盛2006-07-262018-07-092006-07-262018-07-091999http://ntur.lib.ntu.edu.tw//handle/246246/28672細胞免疫在弓蟲感染控制上很重要, 常藉T 細胞分泌各種cytokines 進行。一般 而言,Th1 cytokines 有抑制弓蟲效果,而 Th2 cytokines 卻促使疾病嚴重化。維生素E 為一主要的脂溶性細胞膜抗氧化物,可保 護細胞免受自由基之破壞。另外,維生素E 亦可為免疫調控物質,對維持正常的免疫 功能很重要。維生素E 對於培養的淋巴球 有保護與刺激作用,但原因並非是單純的 由於減低細胞膜之lipid peroxidation。同 時,維生素E 可促使IL-2 及IFN-g之產生, 為Th1 型免疫反應之一種 immunomodulator。T 細胞產生Th1 型 cytokines,已知是受了巨噬細胞分泌之IL- 12 所影響。目前的研究顯示,維生素E 對 維持與促進巨噬細胞功能很重要,如增進 吞噬作用,與IL-1 之分泌。另外,以維生 素E 處理過之巨噬細胞,可增進淋巴球對 mitogens 之反應。由於弓蟲保護性免疫以 Th1 型的細胞免疫反應為主,而維生素E 作用於免疫細胞之機制除了抗氧化之一般 性功能外,亦有助提升Th1 型反應。由此 推知,維生素E 在弓蟲抗原刺激宿主免疫 反應之調控上,應佔有相當重要的角色。 然而,目前有關這方面的報告卻缺如。所 以若能了解維生素E 如何調控抗原誘發之 細胞免疫反應(特別是Th1 型反應),則將 有助發展維生素E 成為一良好的免疫注射 用佐劑。Cell-mediated immunity (CMI) is important in controlling Toxoplasma gondii infection frequently through the release of cytokines by T cells. In general, Th1 cytokines inhibit T. gondii proliferation and Th2 cytokines promote severity of the disease. Vitamin E (VE) is the major lipid soluble membrane antioxidant which protects the cell from the damage by free radicals. VE can act as an immunomodulator which is essential in maintaining normal immunological activity; also, VE can influence intracellular activity, though these mechanisms are not clear. VE can protect and stimulate cultured lymphocytes and this is not simply due to decrease of membrane lipid peroxidation. Also, VE is a Th1- response modulator by inducing IL-2 and IFN-g production. The production of Th1 cytokines by T cells has been known to be affected by IL-12 from macrophages. Current studies have shown that VE is important for maintaining and enhancing macrophage activity by promoting phagocytosis and IL-1 secretion. In addition, VE-treated macrophages enhance lymphocyte response to mitogens. Because Th1-type CMI is the major protective immunity against T. gondii, and because VE promotes Th1 response in addition to its antioxidant function on immune cells, VE should play a significant role in the modulation of immune response stimulated by T. gondii antigens. However, so far no report concerning this matter has been published.
