2011-08-012024-05-18https://scholars.lib.ntu.edu.tw/handle/123456789/706673摘要:2008年國民營養調查顯示,西方飲食是造成國人肥胖的主因。肥胖不僅會引發心血管疾病,心血管疾病死亡率更高居國人死亡的第二位。 因此, 確切了解西方飲食與肥胖及心血管疾病的關連,刻不容緩。 本研究團隊由瘤胃微生物Ruminococcus albus 7萃取出的細菌素albusin B,不僅對格蘭氏陰性病原菌具有抑菌活力,最近的研究更發現此細菌素具有提高腸道抗氧化的能力、調控腸道菌叢組成,及降低血中三酸甘油脂及低密度脂蛋白的功效,這有趣的結果引起我們探討細菌素調控能量代謝相關機制的興趣。 本計畫將以西方飲食(高油脂與高糖份飼糧)餵飼小鼠以誘發肥胖,再以細菌素albusin B治療肥胖的小鼠,藉此探討細菌素對於西方飲食引發的肝臟及心臟能量代謝異常所扮演的角色,作為未來相關研究的依據。此外,本計畫也希望釐清細菌素對於改善能量代謝異常的分子機制,並確認細菌素在治療肥胖引發的代謝疾病的應用。 計劃目的: 1. 建立西方飲食引起肥胖的小鼠模型 2. 釐清健康與肥胖個體二者的心臟與肝臟能量代謝之差異與調控的機制 3. 找出細菌素改善心臟及肝臟代謝異常之可能分子機制 4. 尋找可能參與之蛋白質及其生物活性探索 5. 增加瘤胃微生物產品(albusin B)在臨床上的應用性 <br> Abstract: According to National Nutrition Survey 2008, western diet is the main cause of obesity in Taiwan. Obesity has been demonstrated to induce cardiovascular disease which is the second place of death causes in Taiwan. As a result, to find out the relationship between western diet, obesity and cardiovascular diseases demands immediate attention. Bacteriocin, “albusin B” extracted from the ruminal bacterium R. albus 7 by our research group has the inhibitory activity on gram-negative pathogenic bacteria. In our previous study, it improved the antioxidative ability in the intestine and modulated the gut microbiota. In addition, it decreased the plasma level of triglyceride and low density lipoprotein as well. These interesting results inspired us to study the related mechanisms of bacteriocin on the energy metabolism. In this project, western diet (high fat and high sugar) will be fed to the mice for inducing obesity, then the bacteriocin (albusin B) will be applied to these obese mice. By this approach, the role of bacteriocin on the metabolic dysfunction of liver and heart induced by the western diet will be explored. Moreover, the molecular mechanism of bacteriocin in the improvement of energy metabolism will be elucidated, and the application of bacteriocin on the treatment of obesity-induced metabolic diseases will be confirmed. The major goal for this project is: 1. To establish the mouse model of western diet-induced obesity 2. To elucidate the differences on energy metabolism and molecular regulation of heart and liver between healthy and obese mice 3. To investigate the molecular mechanisms of bacteriocin albusin B on the improvement of metabolic dysfunctions of heart and liver 4. To explore the novel proteins and study their bioactivity 5.
