2012-08-012024-05-13https://scholars.lib.ntu.edu.tw/handle/123456789/642619摘要:(1) 近年來的研究証實「一群具有味覺細胞特性(含有味覺傳導分子 gustducin-alpha、PCL-beta-2 或 味覺受器分子 T1R1、T1R2、T1R3 或 T2Rs 等)的細胞,散布於管腔黏膜 (mucosa) 之表面,或存在於上皮細胞間或位於腸腺或胃腺之細胞間;(2) 這些細胞雖分別經免疫組織化學之研究亦証實胃腸中味覺細胞之其存在及可能含有或釋放某些生物胺(biogenic amine) 如5-HT(serotonin)及特殊之消化激素(hormone) 如gastrin、GLP-1(glucagon-like peptide)、ghrelin 等;(3) 電子顯微鏡研究亦證實:胃腸之味覺相關細胞可能是刷緣細胞或內分泌細胞。依分泌顆粒的特徵、大小和形狀,胃腸道中有許多刷緣細胞及多達18 種以上之腸內分泌細胞(enteroendocrine cells)。目前對胃腸之味覺相關細胞之研究正方興未艾,但大多仍為免疫組織化學與細胞生物學之證據,且其功能大多屬於臆測,缺乏直接之形態學與動物實驗證據可以證實刷緣細胞或何種腸內分泌細胞(1/18)確實為味覺相關細胞 (五種基本味覺中之甜、苦、鮮)。故本計劃擬以兩年時間,擬進行免疫組織化學與免疫電子顯微鏡術(histochemistry, EM andimmune-EM),對成熟小鼠的胃腸的內分泌細胞,進行組織學、超微結構及味覺受器( T1R1、T1R2、T1R3 或 T2Rs) 與腸胃激素之共同定位(colocalization)之研究, 針對:刷緣細胞或何種腸內分泌細胞確實為味覺 (甜、苦、鮮味覺接受器)相關細胞,提出直接之形態學與動物實驗之證據。並以迷走神經切斷術(vagotomy),切斷迷走神經後,對各類型胃腸內分泌細胞的形態改變來推斷可能該等細胞之生理功用。<br> Abstract: Research in recent years confirmed that "a group of taste cells withcharacteristics molecules (i.e., taste transduction molecule, such as gustducin-α,and PCL-β-2 and/or taste receptor T1Rs and T2Rs ) distributed in surfaceepithelium of gastro-intestinal mucosa or among the cells of gastric and intestinalglands. Though these cells were identified by immunohistochemistry, previousstudies confirmed that gastrointestinal taste cells related elements may contain orrelease some biogenic amines (such as 5-HT or serotonin), and digestivehormones (such as gastrin, GLP-1 ( Glucagon-like peptide), GIP, peptide YY andghrelin). Moreover, electron microscopy also revealed that gastrointestinal tastecells related elements may be the brush cells (similar to those in respiratory system)or entero-endocrine cells (up to now, about 18 types were identified).Currently researches on gastrointestinal taste cell related elements areascendant. Most studies were focused on histo-chemistry, physiology, and cellbiology of these cells, and their functions mostly are belonging to speculation andare lacking direct evidence from morphological and animal experiments.In order to determine whether brush cells or which intestinal endocrine cells(1/18) are responsible for taste cell related elements (sweet, bitter, and umamitaste), we propose a two-year project.For the first year, we will identify the gastro-intestinal taste cell related elementsby histochemistry, EM and immune-EM on brush cells and gastrointestinalentero-endocrine cells of mature mice, to reveal the histology, ultrastructure andcolocalization of taste receptor (T1R1and theT1R2, andT1R3 or T2Rs) andgastrointestinal hormones (5-HT, gastrin, GLP-1, GIP, peptide YY and ghrelin).
