2001-08-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/686043摘要:相思樹為台灣常見的本土樹種,主要分布於台灣低海拔山地與丘陵地。目前,有關相思樹的主要功用,除了可供作為傳統薪炭材之外,亦可作為部分結構用材。此外,相思樹為具植物相剋作用之樹種之一,其部份葉部抽出物中所含之Flavonol galloylglycoside類化合物,經海蝦致死試驗已證實具有生物活性。然而,對於相思樹樹皮、心材甚至是葉部中所潛藏具保健醫療功效之抽出物,截至目前為止仍缺乏相關性研究。因此,本研究擬利用管柱層析、高效能液相層析等技術,分離及純化相思樹抽出成分,並對其生物活性、抗氧化與抗腫瘤活性等進行評估。其中生物活性部份,擬利用抗細菌、抗腐朽菌以及抗白蟻試驗等評估相思樹抽出物中具活性之成分;而抗氧化以及抗腫瘤(或癌症化學預防)活性評估,則嘗試利用無定形自由基捕捉、超氧自由基捕捉、脂質過氧化、氫氧自由基引發DNA破壞等體外試驗(In vitro)、細胞抗氧化活性之半活體試驗(Ex vivo)以及動物活體試驗(In vivo)等方法進行檢測。經由上述活性評估所得之具生物活性、抗氧化與抗腫瘤活性成分,則運用質譜分析、紅外光光譜分析以及核磁共振光譜分析等光譜分析,鑑定其確切結構。本計畫<br> Abstract: Acacia confusa Merr. (Leguminosae), a species indigenous to Taiwan, is widely distributed on the hills and lowlands of Taiwan. It represents a traditionally important commodity used for charcoal-making and construction materials. It has been reported that A. confusa is an allelopathic plant, and the leaves contain flavonol galloylglycosides, some of those exhibit anti-hatch activity against brine shrimp. However, the potential health benefits of A. confusa bark, heartwood, and leaf extracts have not been studied to date. In this study, column chromatography (CC) and high performance liquid chromatography (HPLC) were employed to separate and purify the A. confusa extracts, and their bioactivities, antioxidant activities and antitumor activities were further evaluated by various assays. The bioactivities investigated include antifungal, antibacterial and antitermite. As for the antioxidant activity and antitumor activity, both were evaluated using a variety of in vitro, ex vivo, even in vivo model systems, including free radical and superoxide radical scavenging assays, lipid peroxidation assay, hydroxyl radical-induced DNA strand scission assay, cellular antioxidant assay, as well as animal model assay. In addition, the chemical structure of bioactive compounds from A. confusa will be identified by the analyses of mass spectroscopy, Fourier transform infrared spectroscopy (FTIR), and nucleus magnetic resonance spectroscopy (NMR), etc. The accomplishment of this project would help determine the bioactive or antioxidative or antitumor components in the extracts of A. confusa and also elucidate their mechanisms of the inhibitory activity.
