2019-01-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/682366摘要:超廣譜乙內醯胺酶 (extended-spectrum β-lactamases, ESBLs) 可分解乙內醯胺 類抗生素,而這些攜帶有超廣譜乙內醯胺酶的細菌則造成人醫或獸醫治療上的困難 。近年來,具有超廣譜乙內醯胺酶的腸內科細菌,特別是具超廣譜乙內醯胺酶的大 腸桿菌開始在畜禽動物出現,這也造成了人們開始疑慮這些微生物是否可能會藉由 食物鏈傳播至人類。包括豬、雞、牛等肉食動物在台灣的農業產值極具重要性,至 於在這些動物身上是否有攜帶超廣譜乙內醯胺酶的細菌等研究則相對較少。本研究 計畫的目的就是希望能分析由豬、雞、牛等動物分離出來的具超廣譜乙內醯胺酶的 大腸桿菌。簡單來說,從上述動物的糞便檢體所分離之大腸桿菌會先以 CHROMagar呈色培養基初步篩選有可能是具超廣譜乙內醯胺酶的大腸桿菌,接著再以 Clinical and Laboratory Standards Institute (CLSI) 所頒佈的Screening and Confirmatory tests方法進行表現型之確認。再利用聚合酶鏈鎖反應及核酸定序分 析ESBL陽性大腸桿菌是否帶有TEM、SHV或CTX等主要的 bla 基因的型別。多重基因座 序列分型法及 BioNumerics軟體則用來分析這些ESBL陽性大腸桿菌菌株基因型及其 親緣上的關係。由此計畫所獲得之結果可提供台灣肉食動物身上攜帶有超廣譜乙內 醯胺酶大腸桿菌基本的特性資料。 <br> Abstract: Production of extended-spectrumβ-lactamases (ESBLs) confer resistance to the commonly used beta-lactam antimicrobials and ESBL–producing bacteria render treatment difficulty in both human and veterinary medicine. ESBL–producing Enterobacteriaceae , particularly ESBL-producing Escherichia coli isolates, have emerged in livestock recently, which may raise concerns regarding possible transfer of such microorganisms to human through food chain. Food animals, such as swine, chicken, and cattle play an important role in agricultural productions in Taiwan, but investigations regarding the status of ESBL in those animals are limited.The objective of this research is to characterize the ESBL-producing E. coli from swine, chicken, and cattle. Briefly, CHROMagar ESBL will be used to screen potential ESBL-producing E. coli from fecal swab samples of the target animals. Combination disc tests will then be applied to confirm those ESBL-producers according to the guideline of Clinical and Laboratory Standards Institute (CLSI). The major bla genes including TEM, SHV, and CTX will be analyzed by polymerase-chain reaction and DNA sequencing. Genotyping of ESBL-producing E. coli will be performed by multilocus sequence typing and phylogenetic relation of ESBL-producing E. coli strains will be investigated by BioNumerics Software version 7.0.
