2005-08-012024-05-16https://scholars.lib.ntu.edu.tw/handle/123456789/667255摘要:海芋又名馬蹄蓮,為天南星科、馬蹄蓮屬多年生球根花卉,近幾年來深受消費者喜愛,栽培面積持續增加,是國內極具發展潛力之球根花卉。海芋種球若攜帶病毒會影響植株生長與切花品質,因此國內目前已有海芋的組織培養系統,利用所生產的組培苗來培育開花球。但在海芋開花球的養成過程和切花生產期間,皆會遭受多種病毒的危害,嚴重威脅國內的海芋產業。本計畫希望在植物病毒常用的防治策略之外,研究另外一新類型防治方法的效果,即利用基因沉寂(RNA silencing)的機制將入侵的植物病毒RNA加以分解,以達到防治目的。為達成此目的因此將以兩年的時間進行研究,以期能(1)了解sense RNA、antisense RNA和dsRNA對於海芋病毒感染植物能力的影響;(2)了解dsRNA的長度和序列是否會影響其誘發RNA silencing的能力;(3)了解sense RNA、antisense RNA和dsRNA對於已存在植物細胞內的病毒是否具有抑制效果;(4) hairpin RNA對於海芋病毒感染植物能力的影響;(5)了解含有hairpin RNA的大腸桿菌核酸溶液防治海芋病毒的效果;(6)了解以農桿菌媒介之基因短<br> Abstract: Calla lily (Zantedeschia spp.), belonging to the family Araceae, is a perennial bulbous flower and has become popular flower crops in Taiwan. If the tubers of calla lily infected by viruses, the growth and flowering of plants will be severely affected. This is the reason why micropropagation are established for producing virus-free, tissue-cultured seedlings and tubers here. Nevertheless, during production period for tuber and cut flower in field, many viruses still threaten the industry of calla lily in Taiwan. To solve the problem, this project plans to study a new control method that uses the mechanism of RNA silencing to degrade the invading calla-infecting viruses. We want to know the following questions: (1) the influence of sense RNA, antisense RNA and dsRNA on the infection of calla virus to plant; (2) the effect of the length and sequence of dsRNA on trigging RNA silencing; (3) the influence of sense RNA, antisense RNA and dsRNA on virus already existing in plant cell; (4) the influence of hairpin RNA on the infection of calla virus to plant; (5) the effect of hairpin RNA-containing E. coli nucleic acid extracts on the infection of calla virus to plant; and (6) the effect of hairpin RNA produced by Agrobacterium-mediated transient expression assay (agroinfiltration) on the infection of calla virus to plant.
