2013-01-012024-05-14https://scholars.lib.ntu.edu.tw/handle/123456789/655479摘要：香蕉是世界性的重要果樹與糧食作物，也是國際貿易的重要農產品之一。然而香蕉病害種類很多，尤其病毒病害往往成為香蕉產業的重大限制因子，並常藉由無性繁殖體(如吸芽與組織培養苗)做廣泛的傳播，造成摧毀性的影響。為有效杜絕香蕉病毒之危害，防治上首重建立香蕉健康種苗制度，生產並驗證無病毒的香蕉種苗；其次則要預防媒介昆蟲，避免田間再感染。但由於香蕉病毒都有其潛伏期，在種苗階段或田間栽植初期皆不易由外表看出病徵，必需仰賴敏感的病毒檢測技術來做明確且快速的診斷。膠金抗體試劑(colloidal gold-labeled antibody strips)目前被視為是最方便的病毒偵測技術，無需儀器、隨時隨地都可做檢測。國際間重要性的香蕉病毒計有6種，包括香蕉萎縮病毒(Banana bunchy top virus)、胡瓜嵌紋病毒(Cucumber mosaic virus香蕉系統)、香蕉條紋病毒(Banana streak virus)、香蕉苞葉嵌紋病毒(Banana bract mosaic virus)、香蕉輕症嵌紋病毒(Banana mild mosaic virus)與香蕉X病毒(Banana virus X)，本計畫擬針對這些香蕉病毒病害，先研製出其單株抗體（monoclonal antibodies），以ELISA法篩選最佳抗體細胞株(clones)，再逐步製成膠金抗體試劑，最後整合成全套式的檢測試劑套組，提供將來香蕉檢疫與防疫之用。又因香蕉是世界性果樹，未來若予以技轉及商品化，將來有機會做國際行銷。<br> Abstract: Bananas (and plantains) are important fruit and food crops in the world, and they are also important agricultural products in the world trade. Several banana virus diseases transmitted by the vegetative propagations such as suckers and tissue cultures have become tremendous limiting factors for banana industry. Establishment of the healthy nursery systems for production of virus-free seedlings is the main measure to control virus diseases of banana. However, the diagnoses of banana virus diseases were not easy especially when they were latent. The modern diagnostic techniques based on immunology and molecular biology are necessary for the rapid and accurate detections of banana viruses. The colloidal gold-labeled antibody strips are considered to be most convenient detection methods for pathogens, and they could be easily used in the field without instruments and equipments. There are 6 important viruses affecting bananas such as Banana bunchy top virus, Cucumber mosaic virus, Banana streak virus, Banana bract mosaic virus, Banana mild mosaic virus and Banana virus X in the world. This project is dedicated to develop colloidal gold-labeled antibody strips for rapid detections of 6 banana viruses through the preparation of monoclonal antibodies specific against different banana viruses, and finally they will be combined into an integrated diagnostic strip kit to aid the quarantine and control of banana viruses.
