2006-01-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/682443摘要:大波斯菊(Cosmos bipinnatus CAV.),為菊科波斯菊屬植物,近年來因台灣農業的轉型,其已成為重要的景觀綠肥植物。民國93年底,首先於花蓮縣光復鄉,其後於花蓮縣其他各鄉鎮,發現具白色病斑病徵的大波斯菊病株。根據大波斯菊病害文獻的研究顯示,在花蓮地區之病害,疑似為日本及歐美地區已記載報導之大波斯菊白斑病,或稱白色黑穗病(white smut),而本病害過去在台灣並未有正式之記載。本計畫之目的,乃在確認本病害於台灣地區發生的情形,並針對種子及病株樣品,發展快速PCR篩檢偵測技術。本年度,將進一步調查此病害之田間發生情形,並收集不同來源或品種之大波斯菊種子,以及田間疑似罹病株樣品,完成核酸之萃取及品質之確認。此外,將設計一般真菌通用性及 Entyloma 屬專一性PCR引子,並以種子及疑似罹病株純化核酸進行PCR增幅反應,PCR產物經選殖及定序,PCR實驗並以確定罹病株為正對照樣品。結果一旦確認,將建立大波斯菊白斑病PCR快速篩選偵測技術之標準流程,以有效控制此病害的擴散。<br> Abstract: Cosmos (Cosmos bipinnatus CAV.), belonging to Asteraceae, has recently become one of the important green manure recreation plant in Taiwan. In December 2004, symptomatic cosmos plants with whitish spots on the leaves were discovered in Hualien County. A similar disease called cosmos white smut was previously reported in Japan and Europe; however, it has not been officially documented in Taiwan. The goal of the project is to investigate the occurrence of cosmos white smut in Taiwan, and also develop the fast PCR detection technique for screening plant samples including seeds. In this year, we will investigate the occurrence of this disease in Taiwan. Seed samples from different sources or varieties, and a suspected diseased plant will be collected. A commercialized and tested plant genomic DNA extraction protocol will be applied. PCR primers for basidiomycetous fungi and/or specific PCR primers for Entyloma species will be designed for PCR amplification. PCR product will be verified and diseased plant samples with described symptoms will be used as positive control.
