2011-08-012024-05-18https://scholars.lib.ntu.edu.tw/handle/123456789/696540摘要:豬第二型環狀病毒(PCV2)感染為目前對養豬產業衝擊最大的世界性新興豬病之一。本研究室藉由活體外(in vitro)感染方式,已針對單獨PCV2或與PRRS病毒混合感染或在細菌內毒素的協同下,對豬肺泡、脾臟巨噬細胞及單核球和單核球衍生的巨噬細胞功能和病毒複製的影響,以及淋巴細胞對PCV2之感受性和巨噬細胞與淋巴細胞間的交互作用等,進行過一系列研究並有多篇論文發表。在延續舊有基礎下,預計以三年、兩階段方式,進一步探討全身性豬第二型環狀病毒相關疾病(PCVAD)患豬之免疫致病機制,其中包括(1)利用cDNA微陣列晶片、定量聚合酶鏈反應、免疫化學染色及原位聚合酶鏈反應,探討豬隻經人工感染PCV2單一病原後,PCV2於豬隻體內的入侵途徑、複製和分佈位置及其對免疫系統造成的影響;(2)藉由上述資料,進一步建立一個以組織微陣列晶片為基礎的研究模式,並在此模式下進行大規模採樣,以分析於田間在PCV2以外其他傳染性、非傳染性多重輔助因子的影響下,對於田間染患全身性PCVAD豬隻所呈現之淋巴細胞流失、肉芽腫性淋巴腺炎病變及免疫功能下降等之分子免疫致病機制,進行更深入、全面性的探討,期能對豬隻由不顯性PCV2感染發展為全身性PCVAD的致病機制有更清楚的全盤性了解,以作為本病後續的防控基礎。<br> Abstract: Porcine circovirus type 2 (PCV2) infection is one of the major emerging swine diseases to adversely affect the swine industry the most. Our laboratory has done serial in vitro studies to evaluate the effects of PCV2 alone or dual infection with PRRS virus (PRRSV) or in combination with bacterial endotoxin on various functions and viral replication in swine alveolar macrophages, monocytes, and monocyte-derived macrophages. Additionally, the susceptibility of lymphocytes to PCV2 and the effects of singular and dual infection of PCV2 and PRRSV on the interactions between lymphocytes and splenic macrophages were also evaluated. The findings have been published in several papers. On the basis of these previous findings, we plan to further study the immunopathogenesis of the systemic PCV2-associated disease (PCVAD) in pigs. The study consists of the following two major tasks. (1) by the use of cDNA microarray, real time PCR, immunohistochemical staining, and in situ PCR, the location of viral entrance, replication, and distribution as well as changes in the immune system will be thoroughly evaluated in SPF pigs experimentally inoculated with PCV2 alone; (2) based on the findings and the established methodology of the first task, a tissue microarray-based study model will be established. Under this model, a large scale of sample collection will be carried out from field subclinical to late-staged systemic PCVAD-affected pigs. Under the effect of PCV2 and various infectious and non-infectious cofactors, the molecular immunopathogenesis of the lymphoid depletion, granulomatous lymphadenitis, and immunosuppression observed in the field PCVAD-affected pigs will be studied in depth. It is anticipated that the mechanism how the PCV2-infected pigs transform from the subclinical infection to the late stage of systemic PCVAD will be elucidated via this study.
