2011-05-012024-05-13https://scholars.lib.ntu.edu.tw/handle/123456789/643520摘要:梨形鞭毛蟲是一種常見寄生於腸道之寄生蟲,可引起腹瀉疾病之集體爆發。梨形鞭毛蟲的生活史中可以滋養體的形式寄生在小腸,滋養體必須分化成囊體,才能存活於體外並感染新的宿主。梨形鞭毛蟲的生活史已被建立於試管中,然而其致病機制及寄生蟲與宿主交互作用之詳細分子機制仍所知甚少。腸燥症(IBS)是一種常見的腸胃道疾病,消耗全世界很大的醫療成本。目前並沒有針對腸燥症的生物標記或動物模式可供診斷或分析。近來在挪威爆發了一次大規模的梨形鞭毛蟲感染,利用殺蟲藥治療後,仍有10%的病人受到了感染後腸燥症的折磨。我們的團隊中,余佳慧博士將建立一套梨形鞭毛蟲感染導致感染後腸燥症的動物模式,這將會對於尋找治療腸燥症及梨形鞭毛蟲感染的用藥有極大的幫助。過去治療腸燥症的用藥主要是針對抗腹瀉,抗憂鬱,抗焦慮的藥以長期治療其症狀,但它們不一定有效。Metronidazole 常被用來治療梨形鞭毛蟲感染,但是其抗藥性及副作用問題,造成其使用效果不佳。我們實驗室最終目的是要了解梨形鞭毛蟲如何與腸道作用造成危害,並且要尋找更好的治療用藥。中草藥是天然的配方,較可能有較少的副作用,適合做長期治療腸燥症的用藥,也會有很好的市場潛力。已知有幾種草藥可用來治療梨形鞭毛蟲感染,但對於其中的特異性成分和詳細的殺蟲機制仍不清楚。此計劃我們將鑑定新的宿主與梨形鞭毛蟲作用的致病因子,我們將會聚焦於尋找中草藥中更佳的治療梨形鞭毛蟲感染的藥物。如果找到更佳的治療藥物,梨形鞭毛蟲感染的症狀會緩解,並且清除梨形鞭毛蟲也會幫助緩解腸燥症的症狀。<br> Abstract: Giardia lamblia is a common intestinal protozoan parasite responsible for outbreaks ofwaterborne diarrhea. Giardia trophozoites that colonize the upper intestinal tract mustsuccessfully encyst in order to infect a new host. While the entire life cycle can be reproducedin vitro, the pathogenesis and molecular mechanisms involved in host-parasite interaction arepoorly understood. Irritable bowel syndrome (IBS) is one of the most commongastrointestinal disorders leading to high healthcare costs worldwide. Currently, there is nobiological marker and animal models for diagnosis and analysis of IBS, respectively. Anoutbreak of giardiasis occurred in Norway recently. After treatment, 10% people still sufferedfrom post-infectious IBS. In our team, Dr. Linda Chia-Hui Yu is going to establish an animalmodel for Giardia-induced post-infectious IBS, which will be very helpful to find new drugsto treat IBS and giardiasis. Medications for IBS include anti-diarrheal, anti-depressants, oranti-anxiety drugs with long-term management of symptoms, but they are not alwayseffective treatment. Metronidazole has been used often in the treatment of Giardia infection,but resistance and side effect limits its use. In our laboratory, the major final goal is tounderstand how this organism interacts with the host’s intestinal wall and to find an improvedtreatment for Giardia. Chinese medicines and herbs, which are more natural formulations,may have very little side effect, be more suitable for long-term treatment of IBS and have agreat market potential. Several herbs have been used to treat giardiasis. Little is known ofspecific compounds in those herbs and detail mechanisms of their anti-Giardia effect. In thisproject, we will identify new pathogenic factors involved in host- Giardia interaction andpathogenesis. We will focus on Chinese medicines and herbs to find a better treatment ofgiardiasis.
