2011-01-012024-05-14https://scholars.lib.ntu.edu.tw/handle/123456789/659086摘要: 本計畫的目標為針對我國最具國際競爭力的番茄、番椒與青花菜等蔬菜作物,開發簡單重複序列(SSR)及單核?酸多型性(SNP)分子標記系統,配合以具有目標特性之種原所雜交建立的分離族群,建立高密度標記連鎖群圖譜,以定位控制番茄抗晚疫病、番椒雄不稔系統稔性恢復基因、與青花菜早生性之基因座,並將與上述與各作物重要性狀基因緊密連鎖之分子標記,以及具有該基因之衍生育種材料,技術轉移給國內種苗業者或農業研究機構進行分子輔助選種。在第一年的計畫中,已完成 (1) 產生2組青花菜F1雜交種子,以及番椒與番茄各2個F2分離族群;(2)以次世代定序技術所獲得的大量序列資料,探勘獲得約50,000組SSR與700,000組SNP分子標記;(3) 從已公開之文與資料庫,篩選在參試品系間具有多型性的337組番椒與453組青花菜SSR分子標記;(4) 驗證篩選120組探勘獲得之番椒SSR分子標記。本年度的計畫的主要工作項目包括:(1) 建立番茄、番椒各2個F2:3進階分離族群與2個青花菜F2分離族群;(2) 驗證篩選番椒與青花菜各200組探勘所獲得gSSR分子標記;(3) 調查番椒F2族群與F2:3進階分離族群之花粉稔性,以及番茄F2:3進階分離族群之抗病性;(4) 分析番椒與番茄分離族群基因型,建立番椒與番茄連鎖群圖譜。<br> Abstract: Objectives of this research are developing simple sequence repeat (SSR) and single sequence repeat (SNP) markers for three most economically important vegetables, namely tomato, pepper and broccoli, in order to construct high density linkage maps to facilitate mapping quantitative trait loci controlling tomato late blight resistance, pepper sterility restoration, and broccoli earliness. Closely linked flanking markers of the target genes as well as the derived lines containing homozygous target genes may be transferred to the vegetable seed industries in Taiwan for markers assisted selection or introgression. Four main tasks in this first year are: (1) Developing broccoli F<SUB>2</SUB> segregating population and pepper and tomato F<SUB>2:3</SUB> advanced segregating populations, (2) Screening pepper and broccoli markers designed based on the results of mining assembled short sequences using locally adapted elite lines and germplasm, (3) Phenotyping pepper and tomato advanced populations for male-sterility and disease resistance, respectively, (4).
