2009-08-012024-05-18https://scholars.lib.ntu.edu.tw/handle/123456789/705193摘要:已有許多的文獻發表苦瓜具降血糖功效,但補充苦瓜對免疫調節的影響,少有發表而值得探討。已知苦瓜對PPAR 活化與對PGE2 的抑制作用,可能具的抗發炎作用。故本子計畫欲探討苦瓜對免疫功能和發炎反應的影響、抗發炎作用與抑制肥胖導致高血糖的機制、苦瓜的抗發炎與降血糖或血脂作用下動物的免疫反應。研究內容分別為: 第一年 苦瓜與不同萃物之抗發炎功效。 探討苦瓜不同萃物對細胞激素與發炎介質的影響: 1) 以短暫轉染法篩選苦瓜樣品的NF-κB transactivation 活性; 2) 以巨噬細胞株探討對促發炎介質分泌的影響; 3) 以初代腹腔細胞探討苦瓜樣品影響發炎介質分泌的作用濃度; 4) 以致急性發炎動物探討抗發炎作用 第二年 苦瓜與不同萃物之免疫調節功效。 探討苦瓜不同萃物對免疫細胞分泌細胞激素的影響: 1) 對T 細胞株分泌細胞激素的影響; 2) 影響初代脾臟免疫細胞分泌細胞激素的作用濃度; 3) 以BALB/c 小鼠探討對免疫調節的影響; 4) 以卵蛋白致敏小鼠探討對氣喘性過敏免疫反應的影響; 5) 對卵蛋白致敏肥胖小鼠氣喘性過敏免疫反應的影響。 第三年 苦瓜對發炎和免疫調節與脂肪分化調控之交互作用的影響。 以脂肪細胞株培養和高油致肥胖小鼠,探討苦瓜對肥胖高血糖小鼠的抗發炎與抑制脂肪形成或降血糖的相關性。1) 對3T3-L1 細胞株脂肪分化過程介質分泌的影響; 2) 對3T3-L1 和Raw 細胞共同培養下巨噬細胞轉型與脂肪細胞分化的影響; 3)自發性高血糖小鼠的脂肪形成與降血糖的影響; 4)高油致肥胖鼠的巨噬細胞與脂肪細胞分化的影響。 <br> Abstract: Bitter gourd (Momordica Charantia) has been studied for anti-diabetes. However, there is little studied for immunomodulatiory effects on immune responses. Research showed that bitter gourd activated PPAR and suppressed PGE2 production, suggesting its anti-inflammatory effect. Therefore, this project is to evaluate the effect of bitter gourd and components from various fractions, especially water, on inflammation, immune responses, and the interactions between adipocytes and adipose-tissue macrophages in mice with normal and high blood glucose. The experiments will be conducted as following within three years: First year: Evaluation of anti-inflammatory effect of bitter gourd. 1) The NF-κB transactivation activity by transient transfection; 2) The secretions of pro-inflammatory mediators by Raw264.7 macrophage-like cell line; 3) Evaluation of the effective dosages by primary cell culture; 4) Investigation of anti-inflammatory effect by LPS-induced sub-lethal acute inflammation. Second year: Evaluation of immunomodulatory effect of bitter gourd. 1) The secretions of cytokines by T cell lines; 2) Evaluation the effective dosages by primary T cells isolated from BALB/c and C57BL/6 mice; 3) Evaluation of the effect on immune responses in BALB/c mice; 4) Effect on airway hyperreactivity and Th2 responses in ovalbumin-sensitized mice. Third year: Evaluation of anti-inflammatory effect on the interaction between macrophage and adipocytes by bitter gourd. 1) The secretions of cytokines by 3T3-L1 cell lines; 2) Evaluation the effect on the interaction between Raw264.7 and 3T3-L1 cells under co-culture; 3) Investigation of the anti-diabetic effect in mice with spontaneous high blood glucose; 4) Investigation of the anti-diabetic effect in mice with high blood glucose induced by high fat diet.苦瓜發炎免疫調節肥胖脂肪bitter gourdinflammationimmunomodulatiorydiabetic苦瓜之保健產品多樣化研究開發-苦瓜產品的抗發炎與免疫調節功效(1/3)
