2021-08-062024-05-13https://scholars.lib.ntu.edu.tw/handle/123456789/644911鳳梨酵素bromelain於臨床上以被用於緩解手術後及外傷後腫脹、副鼻腔炎等發炎症狀;其於脂質代謝異常之疾病的療效並不清楚。本實驗室近期的研究發現鳳梨酵素bromelain有促進脂質分解來達到減緩脂肪肝的療效;根據這些發現,我們將更進一步利用高血脂apolipoprotein E knockout (apoE KO) mice來測試鳳梨酵素促脂質分解之特性可否可以改善高血脂和抑制粥狀動脈硬化之形成。在此研究計畫中,我們將於apoE KO mice 16週齡時餵食bromelain 4週的時間,之後犧牲小鼠, 抽取血液分離血漿,進行血脂肪分析與發炎細胞激素之分析,心臟主動脈瓣將進行粥狀動脈硬化病變分析血管病變大小與組織免疫染色, 血管將抽取蛋白質以western blot analysis分析血管壁與膽固醇代謝相關的蛋白質表達。此外,我們將利用細胞培養模式來進行bromelain抑制粥狀動脈硬化病變進程之分子機轉探討。最後,本實驗室將與夏建忠博士合作,利用apoE KO mice的模式來測試核子影像醫學於心血管疾病之應用, 並藉由其研究專長來使本計畫之實驗設計與結果之判讀更為精準,有利於此計畫之執行與日後之臨床應用。 Bromelain is clinically used to relieve swelling, paranasal inflammation and other inflammatory symptoms after surgery and trauma; however, its efficacy in the deregulation of lipid metabolism and atherosclerosis is not clear. Recent studies in our laboratory have found that bromelain can promote lipid breakdown to reduce fatty liver. Based on these findings, we will further use hyperlipidemia apolipoprotein E knockout (apoE KO) mice to investigate the effect of bromelain in lipid metabolism and the progression of atherosclerosis. In this research project, apoE KO mice at 16 weeks of age will be fed with bromelain for 4 weeks, then the lipid profile, expression of cytokines, lipid metabolism-related genes and histopathology of atherosclerosis will be analyzed in these mice. Finally, we will work with Dr. Hsia to investigate the protective effect of bromelain on atherosclerosis by using nuclear medicine approach.
