2015-05-012024-05-13https://scholars.lib.ntu.edu.tw/handle/123456789/646869摘要:將外來基因導入哺乳動物的基因組中為生物醫學研究的一大進步,這些基因轉殖動物在基因調控的機制、及其相關研究中提供了許多新的見解和發現。因小鼠的生理構造與人類相似、具多樣性的品系及遺傳背景、發展完備的育種技術,以及蓬勃發展的基因轉殖技術,使得基因轉殖小鼠所扮演的角色日益重要,成為生物醫學領域最常用於探討基因功能與解析人類疾病致病機轉的動物模式。本核心設施的長程目標係以最前瞻的基因剔除/置入(KO/KI)技術,協助使用者研究基因功能、建立人類疾病動物模式、與完成藥物標的的確效驗證;並提供最具效率及最大效益的一站式基因轉殖鼠核心設施,強化學研界與生技產業的連結。主持人自2005 年起承接基因轉殖鼠核心設施(A4)計畫以來,已接受來自101 個研究單位、185 位研究學者申請的基因剔除/置入計畫,至今已協助使用者客製化產出超過252 種可繼代遺傳的基因剔除或置入小鼠。本團隊目前為可提供我國從事生醫研究人員,進行基因剔除/置入鼠技術服務的團隊之一。主持人在本期計劃中整合國立臺灣大學動物資源中心與臺大醫學院實驗動物中心共同參與,期望可同時提供以胚胎幹細胞技術為基礎,及TALEN 與CRISPR/Cas9技術為基礎的基因剔除及基因置入小鼠服務,成為國內生醫研究重要支援單位。<br> Abstract: Recent advances in manipulation of mammalian genome have provided numerous newinsights and findings to expand our understandings in biological and biomedical sciences.Mice has become one of the best animal models for investigating genetic functions with itssimilarity in physiology with human, well-developed breeding knowledge and advancedtransgenic or genetic knockout/knock-in technologies. Thus, genetic modified mice modelswere widely used in biological and medical researches. The objectives of transgenic mousemodels core (TMMC) facility are to provide most-efficient and cost-effective one-stopservices and apply advanced genetic knockout/knock-in technology for establishing micemodel of human diseases, validating efficacy of potential drug targets, and facilitateinternational competitiveness. The principle investigator, Dr. Shu-Wha Lin, has been incharge of the A4-TMMC project since 2005, and have received 384 cases from 185 P.I. over101 research institutes. TMMC facility have produced 252 germline-transmitted knockoutand/or knock-in mice in customization so far. TMMC facility is composed of anexperienced team which is highly skilled in establishing customized genetic knockout andknock-in mice models. For studying gene function specifically, core facility provides theCre-loxP based approach in generating conventional and conditional knockout mice. Inaddition to embryonic stem (ES) cell targeting and microinjection services, core facilityprovides services for targeting vector construction, large-scale Southern blotting screeningof targeted ES cells, and chimera mice breeding. Up to now, TMMC facility has been one ofthe teams capable of providing genetic knockout/knock-in mice production services forresearchers in Taiwan. In cooperation with laboratory animal center of College of Medicineand animal resource center in National Taiwan University, we have a proposal to provideour expertise in comprehensive one-stop services for production of geneticknockout/knock-in mice through ES cell based and genome editing technologies (TALENand CRISPR/Cas9) based approaches.
