2009-08-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/687139摘要:許多彈狀病毒感染人類、牲畜和農作物,威脅到牠們的健康和生產力,其中狂犬病病毒 (Rabies virus) 是最為人熟知的一種。儘管狂犬病病毒不是經由昆蟲傳播,但是許多動物和植物彈狀病毒都必需經由昆蟲媒介。當昆蟲傳播彈狀病毒時,病毒也會在昆蟲體內複製、增殖,所以昆蟲在這裡扮演兩個角色:寄主和病媒。我們的長期目標是瞭解彈狀病毒與其寄主之間的相互作用之分子基礎。Sigma 彈狀病毒 (Sigma rhabdovirus) 僅感染果蠅,所以被視為研究彈狀病毒與寄主的交互作用之理想模型。希望藉由研究模式物種,將結果應用於動、植物彈狀病毒的研究。這個研究計畫的主要目的是找出影響 Sigma 彈狀病毒在黃果蠅 (Drosophila melanogaster) 體內增殖的寄主因子。我們將採集野生黃果蠅以篩選與建立 Sigma 病毒株,並完成 Sigma 病毒完整的基因體定序。然後,進一步建構 Sigma 病毒的感染性選殖株 (infectious clone)。最後,利用酵母菌雙雜合分析 (yeast two-hybrid) 找出與 Sigma 病毒蛋白交互作用的寄主蛋白。RNA 干擾 (RNAi) 試驗將進一步證實,這些寄主基因確實影響 Sigma 病毒在細胞內的複製與形態發生。篩選得到的寄主基因,將是未來發展抗病毒藥物的主要標的。這個計畫所建構的感染性選殖株,可對 Sigma 病毒做反向遺傳學 (reverse genetics) 的研究,並可提供給果蠅研究族群,做為果蠅基因 RNAi 的載體。<br> Abstract: Many rhabdoviruses are economically important pathogens of humans, animals, and plants, of which Rabies virus is the most well known species. Insects transmit many animal and plant rhabdoviruses to their hosts although Rabies virus is not transmitted by vectors. Rhabdoviruses replicate in their insect vectors so insects play two roles in this system: host and vector. Our long-term goal is to understand molecular basis of the interactions between rhabdoviruses and their hosts. Sigma rhabdovirus only infects Drosophila flies, and it provides a unique system to characterize factors governing virus-host interaction. Studying model organism, we hope the results can be applied to animal and plant rhabdovirus research. The aim of this proposed research is to study host regulation of the multiplication of Sigma virus in Drosophila melanogaster. We will screen Sigma virus from natural populations of D. melanogaster and establish virus stocks in the laboratory. We also plan to complete the first full-length genome of Sigma virus and construct infectious clones of Sigma virus. Finally, the use of yeast two-hybrid analysis and RNA interference (RNAi) will identify the host genes affecting replication and morphogenesis of Sigma virus. These host genes are potential targets for the development of antiviral therapeutics. Our Sigma virus infectious clones will be accessible to Drosophila research community.
