2010-08-012024-05-18https://scholars.lib.ntu.edu.tw/handle/123456789/709020摘要:發炎性腸道疾病(Inflammatory bowel disease, IBD) 是常發生在西方國家的慢性及 復發性腸道發炎疾病。益生菌長久以來即被應用於各種發酵食品的製造,除可提供製品 特殊的風味、質地、保存食品外,許多研究指出益生菌具有維持人體腸道內正常微生物 菌相、活化免疫系統、抗氧化性、降低血液中膽固醇含量、抗腫瘤及抗致突變性等諸多 生理功能。有學者認為益生菌的生理功效來自於胞外多醣,益生菌胞外多醣具有益菌助 生性、免疫調節作用、抗突變和抗腫瘤活性。有研究指出IBD 的發生和腸道的氧化壓力 與腸內菌相有很大的相關性。 本研究擬以本研究室過去研究具有較佳雙叉桿菌助生性的益生菌Lactobacillus acidophilus BCRC 14079、L. delbrueckii subsp. bulgaricus BCRC 10696、L. plantarum BCRC 11697、L. rhamnosus GG BCRC 16000 與Lactobacillus casei 01 等五株益生菌胞外 多醣為基質。擬以三年為期,首先評估益生菌胞外多醣對胃酸或膽鹽的耐受性及其對雙 叉桿菌生長的影響。其次,探討益生菌胞外多醣與糞便檢體共培養的菌相變化與丁酸等 代謝產物產量變化情形。最後以動物結腸炎模式去研究益生菌胞外多醣對於大鼠結腸損 傷、腸內菌相及基因表現圖譜的影響。以期闡明益生菌胞外多醣的益生性以及其對結腸 發炎之相關特性。<br> Abstract: Inflammatory bowel disease (IBD) is a group of disorders characterized by a chronic and relapsing inflammation of the gastrointestinal tract frequent in Western countries. IBD is associated with intestinal oxidative stress and microflora. Probiotics are used to make many kinds of fermented foods. They not only provide unique flavor and texture, but also balance intestinal microflora, stimulate immune system, antioxidation, anticarcinogenesis and antimutagenicity. Some researchers consider the healthy benefits of probiotics are related to their polysaccharides, for their prebiotic effect, immunomodulation, antimutation and antitumor activities. Potential protective roles of exopolysaccharides from probiotics against colonic inflammation are highly interested. Several exopolysaccharides from probiotics, which has been studied in our laboratory for their prebiotic effects on bifidobacteria will be studied in this 3 years’project. First, the prebiotic effect of exopolysaccharides from several probiotics on the acidic and bile treatment mimicking gastrointestinal conditions will be established. Second, we will demonstrate that the rate of butyrate formation and the species of butyrate-producing bacterium in the faecal-exopolysaccharide co-culture samples. Finally, via in vivo animal colitis model studies, to determine the effects of exopolysaccharides on the changes of intestinal microflora, colonic inflammation and gene expression profile.
