2008-01-012024-05-15https://scholars.lib.ntu.edu.tw/handle/123456789/662666摘要:具檢疫重要性之次病毒類(viroids)有害生物整合性診斷方法之研發與標準化 檢驗流程之建立本計畫目標為對具檢疫重要性之次病毒建立一整合性診斷鑑定技術及標準化偵測流程;以期對日與俱增的農產品進口,如椰子、馬鈴薯等,所潛藏的次病毒病害如: 馬鈴薯紡錘塊莖次病毒(PTSVd)、椰子cadang-cadang次病毒(CCCVd)等,散播及為害國內其它農作物。由於次病毒不帶有或表現任何蛋白質,無法如其他植物病原微生物發展出以抗體辨識特定蛋白抗原的診斷鑑定方法。相對地,次病毒具有相當穩定的RNA基因體,因此,利用核酸探針或是RT-PCR應可專一、快速且精確的進行偵測分析。結合上述開發的分子探針診斷技術及傳統的病徵觀察及生化分析,預計將可建立起一標準化的次病毒檢疫偵測流程。 <br> Abstract: The goal of this proposal aims to develop an integral approach to diagnose and to assay viroid-infected crops as well as to establish a Standard of Procedure (SOP) for detection of viroids. In particular, the quarantined-viroids, such as Potato tuber spindle viroid (PTSVd), Coconut cadang-cadang viroid (CCCVd) and so on, are emphasized. In contrast to other plant pathogens including viruses, viroids lack any expressed proteins, thereby not available for the development of diagnostic techniques based on antibody-protein recognition. However, the stable existence of the RNA genome of the viroid leads its suitable for nucleic acid-base technologies such as Northern blotting, RT-PCR etc. In combination of these promising molecular probe(s) with traditional bioassays and biochemical analysis, an integral approach will be established for an accuracy, rapid, and standardized detection of viroids.次病毒北方點墨雜交反轉錄-聚合脢連鎖反應viroidNorthern blotRT-PCR植物疫病診斷鑑定技術之研發與應用-具檢疫重要性之次病毒類(viroids)有害生物整合性診斷方法之研發與標準化檢驗流程之建立
