2011-03-302024-05-15https://scholars.lib.ntu.edu.tw/handle/123456789/665992摘要:「翡翠水庫糞生微生物之污染源監測調查與物種評估研究」以微生物來源追蹤 (Microbial Source Tracking,MST)為原則,利用現場採樣及實驗檢測之方式,建立以糞生微生物來源追蹤為主軸之污染源辨識方法。並應用於水資源保護管理及農業廢棄物流佈分析,期望對於翡翠水庫之水資源經營管理有所助益。 我國目前依據指標微生物的培養判斷水體是否受到糞便污染,但此法無法分辨出糞便污染之來源。近年來因生物分析技術的進步發展出微生物追蹤法(Microbial Source Tracking, MST)。微生物追蹤法是確定一個特定汙染來源的過程,如雞、牛、羊、鹿之糞便汙染等。不同物種間,由於其攝食行為及其腸道系統之差異性,造成其腸道環境相異,生長的微生物也會不同。基於此特性,可找出每種物種特定之指標微生物,便可設計出宿主專一性之引子 (host-specific primer),進行宿主專一性聚合&#37238;連鎖反應 (host-specific polymerase chain reaction,簡稱 host-specific PCR) ,便可測量水體中是否有特定物種之糞便污染,進而得知污染的來源。 本團隊預計在不同季節採樣,採樣地點主要分布在翡翠水庫水域8個水質採樣點採樣及其上游集水區北勢溪、逮魚堀溪及金瓜寮溪等共6個自選採樣點、水庫放流水區域1處樣點,總計共15個採樣地點。為評估整體水源區之汙染狀況,將以傳統培養法量測指標微生物之濃度,以宿主專一性聚合&#37238;連鎖反應測量雞、豬、牛、人、鹿等指標。同時檢測其他水質變化指標例如溶氧、溫度、pH、總有機碳等。 本計劃預期建立翡翠水庫及上游集水區之適地性糞生微生物污染指標之檢測方法,並找出糞便污染與指標微生物之關聯,進而評估水體中糞便污染分佈與其物種來源,以利未來水資源管理政策之評估。 <br> Abstract: “Survey on fecal bacteria in Taipei Feitsui Reservoir by MST method” intended to establish a fecal source monitoring method by microbial source tracking (MST). The project would develop the molecular markers and validate the method in field study based on the drainage basin of Taipei Feitsui Reservoir. The source of fecal contamination could compare with agriculture activities and help to improve the water resource management in Taipei Feitsui Reservoir Watershed. MST method could determinate the source of fecal pollution by host-specific primer and host-specific polymerase reaction. For the certain fecal microorganisms are associated with the gastrointestinal tract of particular host, the source of fecal pollution could identify by analyzing certain fecal bacteria by MST method. Therefore, we could develop MST method to differentiate the fecal source included chicken, ruminate, swine, deer, and human in the reservoir. To assess the water quality of the river basin and reservoir, we collected water from different area seasonally and analyze the water characteristics at the sampling time. The sample locations are 6 sites that located on the Beishi River basin, 8 sites in Taipei Feitsui Reservoir, and 1 site in the outlet of reservoir. The microbial pollution would be analyzed by traditional cultivation method and MST method. The chemical characteristics included dissolved oxygen, temperature, pH, and total organic carbon. The goal of this study is to provide MST method based on Feitsui Reservoir and verify the relation of MST and fecal indicator bacteria cultivation method.
