2009-01-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/682570摘要:以感染蘭科植物的蕙蘭輪點病毒(Cymbidium ringspot virus, CymRSV)、菸草脆葉病毒(Tobacco rattle virus, TRV)與番茄輪點病毒(Tomato ringspot virus, ToRSV)為對象,研發專一性引子對和multiplex RT-PCR檢測法,並製備CymRSV及ToRSV的專一性抗血清,以建立進口蘭科植物重要檢疫病毒之檢測系統。今年將執行下列工作:(1)根據序列比對結果設計CymRSV專一性引子對,用以基因選殖,並構築CymRSV表現株。(2)製備CymRSV鞘蛋白的重組蛋白,並用以生產CymRSV抗血清。(3)分析我們所製備之CymRSV抗血清的專一性與檢測效果。(4)嘗試篩選CymRSV引子對,配合mt引子對以建立multiplex RT-PCR的反應條件。<br> Abstract: Cymbidium ringspot virus (CymRSV), Tobacco rattle virus (TRV) and Tomato ringspot virus (ToRSV) which are important quarantine viruses can infect orchid plants. To prevent these viruses from being introduced into Taiwan, this project is going to develop the specific primers and multiplex RT-PCR of CymMV, TRV and ToRSV as well as antisera of CymMV and ToRSV, and thus establish a detection system for important quarantine viruses. The following works will be executed this year. (1) Design the specific primers of CymRSV to clone and construct the expression vector. (2) Produce CymRSV antiserum by purified recombinant capsid protein of CymRSV. (3) Test the specificity and efficiency of CymRSV antiserum.
