2018-08-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/687570摘要:雙萜類化合物(diterpenoids)在植物生長,發育以及防禦上具有重要的地位,而其基因表現具有組織特異性,或是甲基茉莉酸(methyl jasmonate, MeJA)誘導性,這些性質使植物可在不同的逆境,或是生長階段選擇生產需要的雙萜類化合物。因為雙萜化合物具有良好生物活性,包括協助植物毒他作用(allelopathy)、提昇植物對昆蟲的抗食性,或是抑制病原菌的生長,可以說是植物與其他生物互通的關鍵角色,具有重要的生態與生理意義。因為雙萜類化合物對植物來說舉足輕重又有良好經濟價值,雙萜類合成酶(diterpene synthases, diTPSs)即成為植物防禦或是藥物研究中的重要目標。臺灣杉(Taiwania cryptomerioides)為重要的造林樹種,也具有豐富的萜類化合物,再加上其特殊的分類地位,使之成為相當良好的研究目標。本計畫延續自臺灣杉雙萜類合成酶的初步成果,首先進行雙萜類合成酶功能定性 ,接著藉由微距陣晶片分析(microarray analysis)共表現基因群(co-expressed genes),找出調控雙萜類合成酶與其修飾下游修飾酵素,釐清雙萜類生合成途徑以及調控網路,並進一步應用於工業生產,以提昇其在農業管理以及藥物發展的利用價值。<br> Abstract: Diterpenoids are essential for plant growth, development, and defense. Some of diterpenoids biosynthetic genes are regulated in a tissuespecific manner, or under different development stage, and some of them are induced by plant hormone. These particular characteristics help plant to synthesize proper chemical in the right place at the right time. Diterpenoids have good bioactivity, such as the allelochemical, antifeedant, and phytoalexin against the fungal pathogen, so its play a decisive role in plant defense and also having high potential values in medical industry. Therefore, diterpene synthases(diTPSs) are crucial enzymes to study plant defense mechanism and improving production of novel metabolites. Taiwania cryptomerioides is an important plantation species in Taiwan, and great amount of terpenoids were found from its extract. Also, its’ unique taxonomic status provide a good source to study diterpene biosynthesis. In this study, a series of experiment is designed based on the expression pattern of MeJA-induced diTPS genes. By carrying out gene co-expression analysis, the candidate genes involved in regulation of diTPS genes or downstream modification of diterpene structure will be screened, and the functional characterization is further undergoing to establish the firmly biosynthetic pathway.
