2017-07-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/674357摘要:瞭解與環境有交互作用的基因是植物研究重要的目標之一. 大多數的分子生物學研究著重一個基因型及人為製造的突變型, 但瞭解自然界的遺傳變異也是非常重要的, 因為這些變異直接控制了植物在自然環境中的生存. 我們預計: 1) 從阿拉伯芥的1000個基因體中, 找出與環境因子相關的基因. 2) 對這些基因, 研究他們在自然界的分佈. 3) 找出一些關鍵性狀, 並研究這些性狀是否與某些雜草化的阿拉伯芥族群有關.<br> Abstract: Understanding the genes interacting with environments is an important goal in plant biology. While most molecular studies focused on one standard genotype and its artificially generated mutants, it is also crucial to understand natural genetic variation, which is responsible for plants’ adaptation and survival in the real world. This sub-project proposes to study natural genetic variation in the model plant Arabidopsis thaliana, providing this joint proposal with complementary perspectives from the evolution in nature. Using a skill set complementary to most other sub-projects in this proposal, we plan to: 1) Identifying genes associated with local environments. Using bioinformatics and the recently sequenced 1000 Arabidopsis genomes, we will scan the genome systematically and identify genomic regions associated with different environments across the Eurasia continent. 2) Investigating the patterns of spatial distribution and evolutionary history of important genes. Here we will focus on three important traits: flowering time, hypocotyl elongation under different light conditions, and drought tolerance. From the list of candidate genes studied previously, we will investigate their spatial distribution and the timing/location of the origin of novel mutations that contribute to environmental adaptation. 3) We will investigate whether these important traits differ between the recently identified “weedy” and “natural” populations of Arabidopsis and perform genetic mapping to search for candidate genes.
