2002-01-012024-05-14https://scholars.lib.ntu.edu.tw/handle/123456789/658954摘要:本試驗之目的在於飼糧中添加富含二十二碳六烯酸(DHA;44%)的藻類多不飽和脂肪酸,以提高蛋黃中DHA的堆積,生產高DHA機能蛋,期望藉由DHA對肝臟中脂質生合成基因的抑制,減少肝臟中脂質堆積對產蛋性能的影響。餵飼產蛋菜鴨和來航雞以玉米-大豆粕為主的飼糧(ME:2803 Kcal/kg;CP:17.1%;Ca:3.4%)。三組試驗,分別添加0%、0.5%和2%的DHA油。每組產蛋菜鴨和來航雞各10隻(共60隻),試驗為期14天;餵飼添加2﹪奶油飼料,適應一週後進行正式試驗。試驗結果顯示,產蛋菜鴨和來航雞的產蛋率皆維持在80%以上。第一週及第二週之產蛋菜鴨和來航雞蛋重,亦未隨著多不飽和脂肪酸的增加而有顯著差異(p>0.05)。第一週產蛋菜鴨蛋中DHA含量,隨著DHA的增加,分別為0.53%、1.25%和3.56%(p<0.01),來航雞蛋中DHA含量則分別為0.42%、1.12%與2.03%(p<0.01);第二週產蛋菜鴨蛋中DHA含量為0.49%、1.53%與3.26%(p<0.01),來航雞蛋中DHA含量則分別為0.56% 、1.75%和2.97%(p<0.01) ,蛋黃的DHA含量<br> Abstract: The study was designed to utilize an algal DHA oil (44% DHA)to enrich yolk DHA content and produce high DHA eggs. The experiment was also designed to study the effect of dietary DHA on reducing the expression of hepatic lipogenesis-related genes in order to alleviate the negative influence of liver fat deposition on laying performance. The results indicated that egg weight and egg production were not affected by dietary DHA. The yolk DHA content and the liver DHA content of both laying hens and Tsaiya ducks were significantly affected by dietary DHA(p<0.01). Duck SREBP1, SREBP2, FAS and HMGCoA reductase were cloned and sequenced for comparison with other species. The homology was high (82~97%) when compared with other species. DNA fragments of these genes were used as probes to determine the expression of lipogenesis-related genes by northern analysis.
