2012-08-012024-05-18https://scholars.lib.ntu.edu.tw/handle/123456789/706484摘要:最新的國民營養調查顯示,台灣地區總體肥胖盛行率高達17%。肥胖不僅會引發心血管疾病,心血管疾病死亡率更高居國人死亡的第二位。研究也指出,飲食歐美化是造成國人肥胖的主因。因此,精確掌握西方飲食與肥胖及心血管疾病的關連,進而減少飲食引起肥胖的代謝異常,將可維護國人健康,並減少心血管疾病的死亡率。 本研究團隊由瘤胃微生物Ruminococcus albus 7萃取出的細菌素albusin B,對格蘭氏陰性病原菌具有強效的抑菌功效,我們更發現albusin B可提高腸道益菌叢組成、降低血脂及低密度脂蛋白,以及增加肝臟對脂肪的代謝。這個有趣的結果引起我們探討albusinB調控肥胖動物生理代謝的興趣。本計畫將以西式飲食(高油脂與高糖份飼糧)誘發小鼠肥胖,再補充albusin B,藉此探討albusin B在西式飲食引發的肥胖所扮演的角色,本計畫將著重於albusin B調控全身性能量代謝及腸道功能的能力。 計劃預期目標: 1. 建立albusin B改善西式飲食引發能量代謝異常的最適劑量與餵飼期 2. 探討albusin B在肥胖小鼠之能量代謝、腸道功能及食慾的調控方式 3. 利用體外細胞模型,探尋albusin B調控代謝異常可能的作用機轉 4. 增加瘤胃微生物產品(albusin B)在臨床上的應用性 本研究結果將可釐清西式飲食引發肥胖到能量代謝異常過程中,albusin B的角色及作用機制,並增加其在治療肥胖引發的慢性代謝疾病之應用。 <br> Abstract: According to the newest National Nutrition Survey, the prevalence of obesity in Taiwan is 17%. Obesity has been demonstrated to induce cardiovascular disease which is the second place of death causes in Taiwan. The report also points out that western diet is the main cause of obesity in Taiwan. As a result, to master the accurate relationship among western diet, obesity and cardiovascular disease, and further to eliminate the probability of obesity- related metabolic dysfunction induced by western diet not only improves the public health, but also decreases the death caused by cardiovascular diseases. Bacteriocin, “albusin B” extracted from the ruminal bacterium R. albus 7 by our research group has the inhibitory activity on gram-negative pathogenic bacteria. In our previous study, albusin B enhanced the composition of gut probiotic microbiotia, decreased the plasma level of triglyceride and low density lipoprotein, and increased the hepatic lipid metabolism as well. These interesting results inspired us to study the underlined mechanisms of albusin B on the energy metabolism of obese animal model. In this project, western diet (high fat and high sugar) will be fed to the mice to induce obesity, then albusin B will be supplemented to these obese mice. By this approach, the role of albusin B on the obesity induced by western diet will be explored, especially the ability of albusin B on the systemic metabolic dysfunction and intestinal function caused by obesity. The major goal for this project is: 1. To establish the optimal dosage and feeding duration for the albusin B treatment on energy dysfunction of dietary-induced obese mouse 2. To explore the underlined mechanism of albusin B on energy metabolism, intestinal function and satiety of dietary-induced obese mice 3. To investigate the molecular mechanisms of albusin B on the improvement of metabolic dysfunctions of heart and liver by in vitro model 4.
