2020-03-252024-05-15https://scholars.lib.ntu.edu.tw/handle/123456789/666060摘要:玉米赤黴烯酮為鐮刀菌屬黴菌所產生之黴菌毒素,其污染在全世界受到高度重視,台灣因氣候溫暖潮溼非常適合黴菌生長,因此容易產生玉米赤黴烯酮的污染,其所牽涉的層面可從食安、畜牧、環境與公衛議題。國際上對於玉米赤黴烯酮相關的研究主要探討其生殖毒性,鮮少討論其對於生物體的老化以及所誘導之神經毒性。本三年期研究計畫將利用秀麗隱桿線蟲作為活體模式生物,進一步去探討不同濃度且慢性暴露玉米赤黴烯酮可能在生物體內產生的老化以及神經退化性疾病的毒性效應。本研究將分為三個研究目標,研究目標一為探討低濃度慢性暴露玉米赤黴烯酮對線蟲老化的影響及其機制;研究目標二為探討低濃度慢性暴露玉米赤黴烯酮對線蟲在神經退化性疾病上的影響及其機制;研究目標三為探討β-amyrin對慢性暴露玉米赤黴烯酮所造成線蟲之毒性效應的保護作用及機制。本研究之結果將得到相關的毒理科學數據,幫助釐清不同濃度且長期暴露玉米赤黴烯酮在生物體內造成老化或神經退化性疾病相關毒性效應及其調控機制,以及可能的預防與保護方式。研究成果將提供政府決策機關相關科學數據,以增添或修改現有飼料與食品的規定與標準,進而保護台灣人民免於暴露在此黴菌毒素的潛在風險中。<br> Abstract: Zearalenone is a mycotoxin produced by Fusarium fungi. The contamination of zearalenone has been a worldwide highly concerned issue. Zearalenone contamination happens frequently in Taiwan due to the warm and humid weather which is suitable for fungi growth. This involves issues of food safety, livestock, environment, and public health. The zearalenone related research has been focused on reproductive toxicity in organism, however, research of aging and neurodegenerative disease related toxic effects is limited. In this 3 years grant, we propose to investigate the toxic effects of zearalenone on aging and neurodegenerative disease using the nematode Caenorhabditis elegans (C. elegans) as an in vivo model organism. There will be 3 specific aims. For specific aim1, we will investigate the toxic effect of low concentrations and chronic zearalenone exposure on aging and the underlying mechanisms in C. elegans. For specific aim 2, we will investigate the toxic effect of low concentrations and chronic zearalenone exposure on neurodegenerative disease and the underlying mechanisms in C. elegans. For specific aim 3, we will investigate the protective effects of β-amyrin against zearalenone induced toxic effects and the underlying mechanisms in C. elegans. Results from this research will provide related scientific data of zearalenone toxicity and advance our understanding on zearalenone induced toxic effects on aging, neurodegenerative disease, and the underlying mechanisms for possible prevention or protective methods.
