2012-06-152024-05-14https://scholars.lib.ntu.edu.tw/handle/123456789/659496摘要:本計畫依據前一年計畫成果,進一步找尋可與B2M蛋白質搭配,適合用於溫/熱季節母豬配種前生理條件檢測之候選蛋白質,以提升檢測之準確性。獲得之成果將有利於針對特定季節進行母豬繁殖條件監控,預期將目標基因重組蛋白質抗體針對溫/熱季節中不同繁殖能力母豬進行測試,以確認蛋白質組合與母豬繁殖性狀之相關性。 同時並針對B2M蛋白質轉譯後修飾產物進行定序,確認其修飾作用類型,進一步進行單株抗體製備,以提升檢測套組之準確性及便利性。另針對microarray所得差異表現基因群進行SNP探勘、功能性分析及haplotype建立,以提供母豬繁殖性狀育種選拔之標的。本計畫預計完成特定季節(溫/熱季)母豬繁殖性能蛋白質檢驗套組雛形,以便針對不同繁殖性能表現之母豬進行檢測,以提供現場進行配種策略調整;另一方面,預計建立差異表現基因影響繁殖性狀之haplotype,冀能實際與快速的縮短種豬選育時程與提升豬隻繁殖效能。<br> Abstract: This project is going to search for candidate proteins cooperate with B2M protein, which appropriate for the detection of physiological condition of sow in pre-mating stage in warm and hot season, to enhance the accuracy of detection. The results obtained will profit the monitoring of sow reproductive condition before mating in specific seasons. The antibodies produced will be tested to those weaning sows in different seasons and different producing ability to confirm the correlation of this proteins combination and sow reproductive traits. Post translational modification product of B2M protein is going to sequencing to confirm the type of modification, and further preparation of monoclonal antibodies, to enhance the accuracy of the test kits and convenience. The differentially expressed genes derived from microarray analysis are advanced study for SNP discovery, functional analysis and haplotype established to provide the subject of sow reproduction traits in breeding selection.
