2002-01-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/684590摘要:豬的脂肪酸之生合成,主要發生在脂肪組織,並且調控此代謝途徑相關之轉錄因子-固醇調節因子結合蛋白一(SREBP1)也大量表達於此。本研究之目的,在利用富含DHA之藻類DHA產品,添加於豬隻飼糧中,以期生產高DHA機能性豬肉,並調節脂質代謝基因調控。餵飼離乳仔豬含2% DHA油或牛脂飼糧18天,採樣時間分飽食及飢餓組,含2% DHA油飼糧可顯著增加各組織中DHA之含量,其中血漿和肝臟DHA含量增加最多,肌肉中DHA含量亦增加約3倍。北方吸漬法分析則顯示,在肝臟組織中,相對於牛脂處理組,DHA顯著減少SREBP1 mRNA之表現量(p<0.05);ACO mRNA表現方面,只有DHA飽食組有顯著促進其表現(p<0.05);FAS mRNA表現量在各組間無顯著之差異,惟DHA飽食及飢餓組有較低之趨勢。肌肉組織中,DHA飽食及飢餓組顯著促進ACO mRNA之表現量(p<0.05)。脂肪組織中,各處理組SREBP1、FAS及ACO mRNA表現量皆無顯著差異。結果顯示,2% DHA油之添加,可以改變肝臟及肌肉組織中基因mRNA之表現。<br> Abstract: The effect of dietary docosahexaenoic acid on porcine gene expression in vivo Ding, S. Liu, Department of Animal Science, National Taiwan University, Taipei 106, Taiwan, Republic of China The fatty acid de novo synthesis in pig is located mainly in adipose tissue. The Sterol regulatory element binding protein, major transcription factor that regulates fatty acid synthesis, is highly expressed in this tissue too. Weaned, crossbred pigs (30-day-old) were fed either a 2% tallow-added, or a docosahexaenoic acid (DHA) oil-added diet for 18 days. The body weight of the pigs was not affected by different dietary fatty acid (FA) composition. The plasma and liver FA composition reflected the composition of the diet. The adipose tissue and skeletal muscle FA composition only partially reflected the diet, indicating great proportion of the FA in these tissues is from endogenous FA synthesis. Adipocyte determination and differentiation-dependent factor 1 (ADD1) is an important transcription factor for adipocyte differentiation and other aspects of lipid metabolism. It modulates transcription of FA synthase (FAS) to regulate the endogenous FA synthesis in the liver and adipose tissue. The ADD1 mRNA was decreased in the liver of DHA treated pigs as compared with the tallow treated pigs. The diets did not have an effect on the ADD1 mRNA in pig adipose tissue. The result suggests that the great enrichment of liver DHA content inhibits the expression of ADD1 mRNA. Such an effect is similar to what was reported in porcine differentiating adipocytes.
