2016-04-072024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/680936摘要:土肉桂(Cinnamomum osmophloeum Kaneh.)為臺灣重要的本土經濟樹種之一,具有多種化學多態性。其中,肉桂醛型土肉桂(C. osmophloeum ct. cinnamaldehyde)葉子精油含有與大陸菌桂皮(C. cassia)皮油相似的成分,具有許多良好生物活性。然而,肉桂醛型土肉桂葉子精油及抽出成分會隨季節變化而改變其含量,而這成分變動之影響機制仍然不明。因此,本計畫將先分析土肉桂單株林木葉子抽出成分、光合作用速率等性質及植株內變異程度,建構土肉桂植株內穩定且具代表性的葉子採樣方法。然後將以此採樣方法用於未來全年取樣分析野外生長之土肉桂葉子抽出成分組成及其葉子蛋白質體的季節性變化。最後再另行採集額外的野外土肉桂植株葉子,驗證分析其主要抽出成分及蛋白質季節變化,同時以同位素稀釋質譜定量目標蛋白群的絕對表現量,進而結合所有試驗結果,推論土肉桂葉子抽出成分季節性調控因子及變化機制。<br> Abstract: Cinnamomum osmophloeum Kaneh is one of the economically important species in Taiwan and chemical polymorphisms were found in its leaf essential oils. Among them, the main components of the leaf essential oils from C. osmophloeum ct. cinnamaldehyde are similar to that of the bark oils from C. cassia, and are with many excellent bioactivities. However, the chemical components within the essential oils or extractives from C. osmophloeum ct. cinnamaldehyde leaves fluctuate due to the seasonal changes, and the mechanism underlying this seasonal variation of its chemical components is still lacking. The purpose of this proposal is to investigate the seasonal chemical-component variations within the leaves of C. osmophloeum ct. cinnamaldehyde. A leaf collecting method reflecting stable and representative within-tree variation will be identified by the analyses of the extractives and photosynthetic properties of the leaves. This method will be further used to sample all the specimens of other years. A year-round leaf sampling scheme will be arranged in the future to identify the seasonal variations of the extractive components and the proteomes within the leaves of C. osmophloeum trees. Extra samplings of the leaves from additional C. osmophloeum trees will be used to verify and quantify the seasonal variation of specific extractive components and protein expressions. The stable isotope labelling coupled with mass spectroscopy analysis will be used to quantify the absolute abundances of protein expressions.
