2023-08-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/674316年齡增長導致肌肉中蛋白質生合成與降解失恆,同時勞動力上升以及缺乏運動也容易造成肌耐力不足與疲勞狀況,導致生活品質變差,連帶增加相關治療和照護費用。前人的研究指出補充一些蛋白質跟胺基酸類機能性食品可以幫助肌纖維之修復、增加肌肉量與肌耐力。屠宰雞隻後的副產物雞肝中含有約17%的蛋白質,且含有動物性產品缺乏的維他命C與硒,而本實驗室開發出的機能性白肉雞肝水解物(chicken-liver hydrolysates, CLHs)富含抗氧化的雙胜肽(肌肽、甲肌肽)、游離態的支鏈胺基酸、組胺酸、精胺酸、牛磺酸等。由於甲基脫氫皮質固醇(dexamethasone, Dex.) 造成肌肉中蛋白質合成與降解失衡、粒線體功能失調之狀況,因此在預實驗發現利用Dex.誘導小鼠肌肉萎縮模式來模擬老化情況,CLHs補充可以增加後肢腓腸肌纖維橫切面積。因此本計畫預計先藉由肌肉細胞(C2C12 cell)試驗評估雞肝水解物(CLHs)對Dex.處理的肌肉中蛋白質合成與降解恆定的影響,希望此細胞試驗結果可供未來動物試驗或人體試驗的依據、亦提供國人未來保健產品的另一個選擇。 Aging results in disturbance of protein synthesis and breakdown in muscles; meanwhile, increasing labor and lack of exercise also lead less muscular endurance and fatigue. Muscle atrophy, less muscular endurance, and fatigue cause poor life quality and then increase cost of therapy and medical care. It has been proven that some protein and amino-acid supplements own abilities to restore muscle fibers, and enhance muscle mass and endurance. Chicken livers, an edible byproduct after slaughtering, contain approximately 17% protein, and vitamin C and selenium which are deficient in animal products. Our research team has been developed functional chicken-liver hydrolysates (CLHs) which contain imidazole-ring dipeptides (carnosine/anserine) and plenty amount of free amino acids, especially branched chain amino acids, histidine, arginine, taurine etc. Because dexamethasone (Dex.) could induce dysfunctions of protein synthesis/breakdown and mitochondrial function in muscles. Based on results from our preliminary experiment, CLHs could enlarge the cross area of hindlimb gastrocnemius in Dex. treated mice. Hence, Dex. will be used to mimic ageing processing which cause muscle atrophy. Then the objective of this study will be to verify if our functional CLHs can promote muscle growth and improve muscular endurance. The current study will try to understand the effects of CLHs on protein synthesis and breakdown in muscle cells (C2C12 cell).
