本計畫首先希透過擴大臨床上常見致病基因在不同病程階段之病患代謝體檢驗,釐清代謝體反映眼底病程變化之代表性;再透過基因轉殖鼠動物模型,探索各階段視網膜神經元單細胞轉錄組的基因表現與同步血清代謝體的連動情形;進而將上述資訊結合我們目前已知之基因型-表現型關聯性分析,達成多體學:基因體-轉錄組-代謝體-表現型整合分析模型,目標期能達成 (1) 擷取重要之代謝體生物標記,發展針對「高風險族群之早期偵測」與「已發病患者之病情監控」可用之快捷代謝體質譜檢測;(2) 揭露個別基因或亞型中重要之生化退化路徑,探索小分子藥物介入之可能性,達成新藥開發之契機,增加整體病患可接受治療之機會。
