Description
此計畫欲建立利用adenine造成腎小管間質傷害的慢性腎病模式,探討在疾病模式中,內皮細胞活化與發炎反應的變化。利用基因剔除小鼠,分別在所有細胞及腎小管細胞剔除angiopoietin-2,欲探討adenine慢性腎病模式,對腎臟纖維化,發炎反應,微血管稀薄化的影響;此外藉由在內皮細胞剔除angiopoietin-2,可探討在單側輸尿管阻塞的漸進性腎臟纖維化模式中,對內皮細胞活化及發炎反應的影響。最後利用細胞分析及細胞實驗,確認致病機制。我們相信,藉由抑制angiopoietin-2,可影響血管內皮細胞活化及發炎反應,發展出慢性腎病治療的新契機。
