The microfluidic device is resealable for cell culture and immunostaining process. Besides, flow conditions, such as flow rates, flow directions and in a single channel can be well-controlled to stimulate a large number of primary cilia. Moreover, the microfluidic platform can directly change different fluids using a three-way valves, which enables the direct primary cilia fixation without stopping the fluid flow. To proof-of-concept, we apply various flow rates flow to stimulate the primary cilium and observe its bending phenomenon. Furthermore, we immunostained over 100 primary cilia in each flow rate to perform a large number bending behavior analysis. In summary, our system is capable of bending multiple primary cilia at the same time and detachable after flow experiment for further analysis. This microfluidic platform presents the ability to stimulate primary cilia and can be integrated with other technique like superresolution microscopy and protein mass spectrometry to study how fluid flow affect both the external structure and the inner biological responses.
