The spheroids were truly spherical and had organotypic densities. The array pitch impacted growth kinetics and was used to tune the spheroid diameter from 200 to 550 μm. During sustained culture mass transfer gradients were established, reflecting the microenvironment of avascularised tumours. The spheroids have reduced chemosensitivity towards the anti-cancer agent irinotecan, with further reduced sensitivity following continued growth into a hypoxic phase of culture. In summary, microarray-based spheroid culture has great potential to support drug discovery and tumour biology research. |
