Description
家族性主動脈瘤暨剝離(TAAD)的預後不良,藥物治療僅能稍微減緩進展,高風險患者仍需手術介入,然而術後嚴重併發症的比例高,需要更深入了解病理機制,以開發相關藥物治療。我們針對家族性TAAD患者進行基因分析,發現一個新型的SMAD3 A34T/+變異會引起胸主動脈瘤快速進展,我們發現SMAD3 A34T並不直接會導致主動脈平滑肌細胞凋亡,而是引起纖維母細胞中非典型TGFβ信號途徑過度活化,是導致TAAD發生的重要病理機轉。本計畫的目的在深入研究SMAD3 A34T突變在主動脈纖維母細胞中誘發TAAD的分子機制,建立相對應的小鼠模型, 並藉此開發TAAD的新型治療策略。
